Then added was tri-o-tolylphosphine (4.92 g, 0.16 mol) then sodium t-butoxide (53.8 g, 0.56 mol) then Boc-piperazine (86.8 g, 0.47 mol) then a solution of 7-bromo-2,3-dihydro-benzofuran (79.6 g, 0.40 mol, prepared according to Tetrahedron Lett. 1998, 39, 2219) in toluene (100 ml). The reaction mixture was stirred at reflux for 16 h. By TLC, all starting material had been consumed. The cooled reaction mixture was filtered over a pad of Celite. The filtrate was concentrated under reduced pressure and the residue was triturated with ethyl acetate in heptane (50%). The insoluble material was filtered off and that filtrate was concentrated under reduced pressure. The crude residue was purified by flash column chromatography using ethyl acetate in heptane (50%) to give 46.4 g (38% yield) of the first intermediate compound as a tan solid., 206347-30-0

Example 16 3-(6-Amino-3H-isobenzofuran-1-ylidene)-5-chloro-1,3-dihydro-indol-2-one To a solution of 5-chlorooxindole (0.629 g, 3.78 mmol) in 10.0 mL monoglyme was added 7.51 mL sodium hexamethyldisilazane (1.0 M in THF) over 3 min. After stirring at room temperature for 8 min, a slurry of 6-aminophthalide (0.561 g, 3.78 mmol) in 4.0 mL of monoglyme was added in one portion. The mixture was stirred for 40 min and then quenched into 100 mL of 4% aqueous HCl solution. The yellow solid was filtered and then partitioned between EtOAc and saturated NaHCO3 (heated to dissolve the solid). The organic phase was washed with H2O, brine and then dried with Na2SO4. The solvent was removed in vacuo and the residue was triturated with MeOH to give the title compound (439 mg, 39%) as a yellow solid.

General procedure: Isobenzofuran-1(3H)-one (0.4 mmol), KOH (0.4 mmol), PhSiH3 (1.2 mmol), dried THF (2 mL) were charged in a Schlenk tube(15 mL) under N2 atmosphere. The mixture was refluxed for 4 h. After cooling to rt, the reaction was quenched with EtOH(0.5-1 mL). Solvent was removed under reduced pressure and the crude residue was purified by column chromatography on silica gel with petroleum ether-ethyl acetate as the eluent to afford the desired product.

Commercially available ethyl 5-NITROBENZOFURAN-2-CARBOXYLATE 30 was HYDROGENATED on PD-C and ACYLATED using commercially available 4-NITRO-N-METHYL-2- TRICHLOROACETYL-PYRROLE 32 to form 33. HYDROGENATION of 33 followed by acylation with 32 afforded the nitro compound 35, which upon HYDROGENATION yielded 36 as the amino compound. ACETYLATION of the amino group of 36 followed by base catalyzed hydrolysis resulted in the acid 37. Coupling of 37 with the hydrochloride salt 39 (obtained by DEPROTECTION of the BOC derivative 38) using EDC afforded the ester 40. See Figure 4.
