General procedure: Isobenzofuran-1(3H)-one (0.4 mmol), KOH (0.4 mmol), PhSiH3 (1.2 mmol), dried THF (2 mL) were charged in a Schlenk tube(15 mL) under N2 atmosphere. The mixture was refluxed for 4 h. After cooling to rt, the reaction was quenched with EtOH(0.5-1 mL). Solvent was removed under reduced pressure and the crude residue was purified by column chromatography on silica gel with petroleum ether-ethyl acetate as the eluent to afford the desired product.

Commercially available ethyl 5-NITROBENZOFURAN-2-CARBOXYLATE 30 was HYDROGENATED on PD-C and ACYLATED using commercially available 4-NITRO-N-METHYL-2- TRICHLOROACETYL-PYRROLE 32 to form 33. HYDROGENATION of 33 followed by acylation with 32 afforded the nitro compound 35, which upon HYDROGENATION yielded 36 as the amino compound. ACETYLATION of the amino group of 36 followed by base catalyzed hydrolysis resulted in the acid 37. Coupling of 37 with the hydrochloride salt 39 (obtained by DEPROTECTION of the BOC derivative 38) using EDC afforded the ester 40. See Figure 4.

Thetriflate ester, compound 16, was produced in 69% yield from compound 15 by reacting it with Nphenyl-bis(trifluoromethanesulfonimide) (1.0 equivalent) in methylene chloride for 72 hours.Compound 16 in a mixture of DMF, methanol, and triethylamine, was added to a preparedsolution of palladium acetate, 1,3-Bis(diphenylphosphino)propane (dppp), DMF and methanol inan autoclave. Carbon monoxide was charged into the autoclave to a pressure of 8 bar, and the reaction mixture was heated at 70 C for 6 hours. After workup, compound 17 was isolated in 91% yield. Lithium hydroxide (4 equivalents) in methanol and water was used to hydrolyze the ester and permit the isolation of compound 18? in 97% yield.

After cooling the mixture, the liberated product was collected by filtration and then washed with water. The obtained pale brown solid was dissolved in ethyl acetate, then the insoluble material was removed by filtration, and the filtrate was washed successively with a 1N aqueous solution of hydrochloric acid, an aqueous solution of sodium hydrogen carbonate, and an aqueous solution of sodium chloride, and then dried over anhydrous magnesium sulfate. After filtration, the filtrate was concentrated to afford the title compound (3.57 g, 84% yield) as a crystalline solid.
