Eleven polyphenolic compounds, including procatechuic and chlorogenic acids, (+)-dihydroquercetin, rutin, isoquercitrin, quercitrin, (+)-dihydrokaempferol, trans-resveratrol, moracin, quercetin and 4-prenylmoracin were isolated and purified from the methanolic extract of defatted mulberry seed residue by a series of column chromatog. including silica gel, Sephadex LH-20, and ODS-A, and their chem. structures were identified by spectral anal. The antioxidant activities of the eleven isolated polyphenolic compounds were measured spectrophotometrically using DPPH radical. Among the eleven polyphenolic compounds tested, rutin (IC50=20.2 μM), isoquercitrin (IC50=22.5 μM), quercitrin (IC50=24.6 μM), quercetin (IC50=27.8 μM), (+)-dihydroquercetin (IC50=28.9 μM), and chlorogenic acid (IC50=30.6 μM) exhibited stronger antioxidant activity than L-ascorbic acid (IC50=31.5 μM) and α-tocopherol (IC50=52.3 μM), whereas procatechuic acid (IC50=68.2 μM) showed lower activity. In addition, (+)-dihydrokaempferol (IC50=33.8 μM), trans-resveratrol (IC50=36.2 μM), moracin (IC50=47.6 μM), and 4-prenylmoracin (IC50=48.2 μM) exhibited moderate antioxidant activity. Furthermore, levels of the eleven polyphenolic compounds from three different types of mulberry seeds were quantified by HPLC, and their contents were as follows: rutin (31.1-60.0 mg/100 g)> quercitrin (7.2-34.2 mg/100 g)> (+)-dihydroquercetin (13.2-33.1 mg/100 g)> quercetin (15.8-19.5 mg/100 g)> 4-prenylmoracin (10.5-43.3 mg/100 g)> isoquercitrin (5.8-15.4 mg/100 g)> chlorogenic acid (0.0-15.3 mg/100 g)> moracin (4.7-7.2 mg/100 g)> procatechuic acid (0.0-11.6 mg/100 g)> (+)-dihydrokaempferol and trans-resveratrol (<0.1 mg/100 g). The ‘Daesungppong’ mulberry seeds among the three cultivars had higher flavonoid contents, such as rutin and quercetin derivatives, while the ‘Iksuppong’ seeds had the highest contents of phenolic acids and moracin derivatives ‘Cheongilppong’ had lower amounts of polyphenolic compounds than the other two mulberry seeds. These results indicate that mulberry seeds containing antioxidant polyphenolic compounds may be potentially useful sources of anti-diabetic, anti-hypertensive, and anti-aging agents for functional foods and cosmetics.

The leaves of Morus alba (Moraceae) have been traditionally used for the treatment of metabolic diseases including diabetes and hyperlipidemia. Thus, inhibitory effect of M. alba leaves on pancreatic lipase and their active constituents were investigated in this study. Twenty phenolic compounds including ten flavonoids, eight benzofurans, one stilbene and one chalcones were isolated from the leaves of M. alba. Among the isolated compounds, morachalcone A (20) exerted strong pancreatic lipase inhibition with IC50 value of 6.2 μM. Other phenolic compounds containing a prenyl group showed moderate pancreatic lipase inhibition with IC50 value of <50 μM. Next, extraction conditions with maximum pancreatic lipase inhibition and phenolic content were optimized using response surface methodol. with three-level-three-factor Box-Behnken design. Our results suggested the optimized extraction condition for maximum pancreatic lipase inhibition and phenolic content as ethanol concentration of 74.9%; temperature 57.4 °C and sample/solvent ratio, 1/10. The pancreatic lipase inhibition and total phenolic content under optimized condition were found to be 58.5% and 26.2 μg GAE (gallic acid equivalent)/mg extract, resp., which were well matched with the predicted value.

Hypoxia-inducible factor-1 (HIF-1) is the central mediator of cellular responses to low oxygen concentrations and vital to many aspects of cancer biol. Bioassay-guided fractionation of the chloroform-soluble extracts of Morus species using a hypoxia response element (HRE)-dependent reporter assay led to identification of six benzofurans (1-6) and two chalcone-derived Diels-Alder adducts (7, 8) from Mori Cortex Radicis and three prenylated benzofurans (9-11) and four chalcone-derived Diels-Alder adducts (12-15) from Morus bombycis. The structure of the new 2-arylbenzofuran-type compound, moracin Q (3), was elucidated by spectroscopic methods, and the absolute configuration of 2 was determined for the first time. The selected compounds (1-3, 5, 7, 9, 10, and 12) from the cell-based reporter assay were found to inhibit hypoxia-induced HIF-1α accumulation in a dose-dependent manner in human hepatocelluar carcinoma cell-line Hep3B cells. Furthermore, these compounds were also active against hypoxia-induced vascular endothelial growth factor (VEGF) secretion in Hep3B cells.

Sulfur (VI) fluoride exchange (SuFEx) reactions can be applied not only in organic click synthesis, but also in the preparation of functional main-chain polymers. In this work, four functional main-chain polysulfates (PNT-PS, NPNT-PS, PHF-PS, and TPE-PS) are synthesized in high yield using the SuFEx reaction at room temperature The polysulfates exhibit satisfactory thermal stability and solution processability. They are used as the active layer for memory devices (ITO/PNT-PS/Al, ITO/NPNT-PS/Al, ITO/PHF-PS/Al, and ITO/TPE-PS/Al). I-V measurements show that ITO/PNT-PS/Al and ITO/NPNT-PS/Al exhibit stable flash-memory (write-read-erase) behavior, while ITO/PHF-PS/Al and ITO/TPE-PS/Al exhibit WORM (write once read many) behavior.
