The relation between colour and structural changes in thermochromic systems comprising crystal violet lactone, bisphenol A, and tetradecanol

The aim of the paper was to find a direct connection between dynamic colour changes, phase changes and chemical interactions in model three-component leuco dye based thermochromic systems. The model systems, containing crystal violet lactone as a colour former, bisphenol A as a developer and 1-tetradecanol as a co-solvent, were analysed by DSC and FTIR spectroscopy and the results were related to the characteristics of the dynamic colour change. The ternary thermochromic systems were also compared with binary mixtures of the co-solvent with the developer and colour former, respectively. The temperatures characterizing the dynamic colour change at decolouration limits were directly related to the solid?liquid transition on heating and liquid?solid transition on cooling, regardless the concentration of bisphenol A. In ternary thermochromic systems, an indistinctive phase transition at the temperatures below the solid?solid (crystal?rotator) transition was observed. The straight connection between the phase transitions and temperatures characterizing the dynamic colour change at colouration limits was not proved. The colour contrast of thermochromic systems was found to be directly related to the ratio of integrated intensity of lactone ring opened (solid) and lactone ring closed (liquid) carbonyl vibration characterized by infrared spectroscopy.

The structure of the regioisomers was assigned through 2D NMR 1H-13C HMBC experiments performed on 10a and 14a. Compounds 10a,b, 14a as well as phenol 1 and the starting quinones 2, 5, 7 and 15 are evaluated against Leishmania sp., Toxoplasma gondii and THP-1 cells. Almost all the tested compounds exhibit significant antiprotozoal activities with lower cytotoxicities than the reference compounds. Among them, quinones 2 and 14a possess the best activities towards L. donovani and T. gondii with the lowest toxicities.

We report an extensive ?heteroarene scan? of triazine nitrile ligands of the cysteine protease human cathepsin L (hCatL) to investigate pi-stacking on the peptide amide bond Gly67?Gly68 at the entrance of the S3 pocket. This heteroarene???peptide bond stacking was supported by a co-crystal structure of an imidazopyridine ligand with hCatL. Inhibitory constants (Ki) are strongly influenced by the diverse nature of the heterocycles and specific interactions with the local environment of the S3 pocket. Binding affinities vary by three orders of magnitude. All heteroaromatic ligands feature enhanced binding by comparison with hydrocarbon analogues. Predicted energetic contributions from the orientation of the local dipole moments of heteroarene and peptide bond could not be confirmed. Binding of benzothienyl (Ki=4 nm) and benzothiazolyl (Ki=17 nm) ligands was enhanced by intermolecular C?S???O=C interactions (chalcogen bonding) with the backbone C=O of Asn66 in the S3 pocket. The ligands were also tested for the related enzyme rhodesain.

Design, synthesis and Structure-activity relationship studies of new thiazole-based free fatty acid receptor 1 agonists for the treatment of type 2 diabetes

The free fatty acid receptor 1 (FFA1/GPR40) has attracted interest as a novel target for the treatment of type 2 diabetes. Several series of FFA1 agonists including TAK-875, the most advanced compound terminated in phase III studies due to concerns about liver toxicity, have been hampered by relatively high molecular weight and lipophilicity. Aiming to develop potent FFA1 agonists with low risk of liver toxicity by decreasing the lipophilicity, the middle phenyl of TAK-875 was replaced by 11 polar five-membered heteroaromatics. Subsequently, systematic exploration of SAR and application of molecular modeling, leads to the identification of compound 44, which was an excellent FFA1 agonist with robustly hypoglycemic effect both in normal and type 2 diabetic mice, low risks of hypoglycemia and liver toxicity even at the twice molar dose of TAK-875. Meanwhile, two important findings were noted. First, the methyl group in our thiazole series occupied a small hydrophobic subpocket which had no interactions with TAK-875. Furthermore, the agonistic activity revealed a good correlation with the dihedral angle between thiazole core and the terminal benzene ring. These results promote the understanding of ligand-binding pocket and might help to design more promising FFA1 agonists.

Colorimetric analysis of thermochromic samples in different forms employing a digital camera

The calibrated and characterised digital camera was used for the recording of a dynamic colour change of thermally treated three-component leuco dye based thermochromic samples in various forms ? bulk system, dry pigment, water- and solvent-based ink formulations, and their dry layers coated on different paper substrates. The study has proved that the proposed approach enables to examine the thermochromic behaviour through the individual stages of the thermochromic coating preparation process. The measured path in CIELAB colour space and derived characteristics of the dynamic colour change were obtained with higher time-temperature resolution than with previously employed methods, enabling to observe even the less distinct features. The most significant differences were related to the length and complexity of the colour path. Dry layers of the water-based thermochromic ink coated on papers had higher values of the total length of the path and the total colour contrast than the layers of the solvent-based ink. The colour of water-based coatings had higher chroma and also it was less influenced by paper substrate. When compared to the bulk thermochromic system, the dry pigment and dry ink layers on paper substrates exhibited a larger width of a hysteresis loop at the decolouration limit, mainly due to the decrease of the temperature at which the colouration starts on cooling.
