Benzofuran-isatin hybrids and their in vitro anti-mycobacterial activities against multi-drug resistant Mycobacterium tuberculosis

A series of benzofuran-isatin hybrids 6a-n and 7a-g linked by alkyl linkers were designed and synthesized. Among them, hybrids 6a-1 and 7a-g were assessed for their in vitro anti-mycobacterial activities against two multi-drug resistant Mycobacterium tuberculosis (MDR-MTB) strains and the cytotoxicity towards CHO cells. The preliminary results indicated that all hybrids (MIC: 0.125-16 mu g/mL) showed excellent activity against the tested MDR-MTB strains, and low cytotoxicity (CC50: 64->512 mu g/mL) towards CHO cells. Among them, hybrid 7e (MIC: 0.125 and 0.25 mu g/mL) was highly active against the tested two MDR-MTB strains, which was 8-16 folds better than ciprofloxacin (MIC: 1 and 4 mu g/mL), >= 512 folds more potent than rifampicin (MIC: 64 and > 128 mu g/mL) and isoniazid (MIC: >128 mu g/mL), but it was less active than TAM16 (MIC: <0.06 mu g/mL). Moreover, the hybrid 7e (CC50: 128 mu g/mL) also showed low cytotoxicity towards CHO cells, and high selectivity index (1,024). However, the metabolic stability and in vivo pharmacokinetic profiles of hybrid 7e were inferior to TAM16, so it still needs to be modified so as to get the optimized hybrid for potential use in mycobacterial treatment. Synthesis and Anticancer Activity of New ((Furan-2-yl)-1,3,4-thiadiazolyl)-1,3,4-oxadiazole Acyclic Sugar Derivatives

New sugar hydrazones incorporating furan and/or 1,3,4-thiadiazole ring systems were synthesized by reaction of the corresponding hydrazide with different aldose sugars. Heterocyclization of the formed hydra zones afforded the derived acyclic nucleoside analogues possessing the 1,3,4-oxadiazoline as modified nucleobase via acetylation followed by the heterocyclization process. The anticancer activity of the synthesized compounds was studied against human liver carcinoma cell (HepG-2) and at human normal retina pigmented epithelium cells (RPE-1). High activities were revealed by compounds 3, 12 and 14 with IC50 values near to that of the reference drug doxorubicin.

The present study aims to investigate whether (3-(7-methoxy-2-p-tolyl benzofuran-5-yl) propan-1-ol) (DK-1108), new synthetic benzofuran compound exerts anti-asthmatic effects in vitro and in vivo. DK-1108 strongly reduced the production of inflammatory mediators, cytokines and chemokines in RAW264.7 and A549 cells. DK-1108 significantly regulated the levels of AKT/MAPKs/c-Jun activation, AP-1 luciferase activity and ICAM-1 expression. Furthermore, DK-1108 effectively suppressed the adhesion of A549 and EOL-1 cells. In OVA-induced asthmatic mice, DK-1108 decreased the levels of IL-5/IL-13/IgE production, eosinophils/macrophages influx, ICAM-1/MCP-1 expression, mucus secretion and airway hyperresponsiveness (AHR). These effects of DK-1108 were accompanied by downregulation of MAPKs activation. Therefore, we suggest that DK-1108 exerts protective effect against airway inflammation and mucus overproduction, and therefore could be valuable therapeutic agent for treatment in asthma.

Transition metal-catalyzed C-H activation and radical reactions are two versatile strategies to construct diverse organic skeletons. Here we show the construction of a class of flavylium fluorophores via the merge of radical chemistry and C-H activation starting from (hetero)aryl ketones and alkynes. This protocol is not only applicable to aryl ketones but also to heteroaryl ketones such as thiophene, benzothiophene and benzofuran, thus leading to structural diversity. Mechanism studies, including control experiments, intermediate separation, radical trapping, EPR and ESI-HRMS experiments, demonstrate that the key step lies in the addition of the acyl radical generated by the copper-catalyzed C-C bond cleavage of aryl ketone to the rhodacycle formed via the C-H activation of aryl ketone. The flavylium fluorophores feature butterfly symmetrical configuration, nearly planar skeleton and delocalized positive charge, and exhibit intriguing photophysical properties, such as tunable absorption and emission wavelengths and high quantum yields.

The chemical structure of the prepared compound was determined by the elemental analysis and spectral data. The individual emulation characteristics of compound (2, MOFCA), were accomplished by DFT, and TD-DFT/B3LYP, at 6-311 ++ G (d, p). The computational results detect the most stable structure of MOFCA, depending on the positions of the methoxy (O-CH 3) group, within change in dihedral angle. FT-IR spectroscopy was applied for the vibrational spectral analysis. Using frontier molecular orbital (FMO) analysis, various spectroscopic and quantum chemical parameters are discussed. The absorption energies, oscillator strength, and electronic transitions of compound (2, MOFCA), have been derived at TD-DFT/CAM-B3LYP/6-311++ G (d,p) computations utilizing a PCM and measured in different polar and non-polar solvents experimentally in UV-Vis spectra. The output of the computation shows accurate agreement between theoretical spectra and practical spectra for the title compound. NLO analysis was computed at the identical plane of theory which are, alpha; Delta alpha, and first-order , the hyper-Rayleigh scattering (beta HRS) and the depolarization ratio (DR), were shown promising optical properties. The plots of natural bonding orbital (NBO), thermochemical parameters and the molecular electrostatic potential surfaces (MEPS) have been computed. Biomimetic Oxidation of Benzofurans with Hydrogen Peroxide Catalyzed by Mn(III) Porphyrins

The modelling of metabolic activation of the benzofuran nucleus is important to obtain eco-sustainable degradation methods and to understand the related mechanisms. The present work reports the catalytic oxidation of benzofuran, 2-methylbenzofuran, and 3-methylbenzofuran by hydrogen peroxide, at room temperature, in the presence of different Mn(III) porphyrins as models of cytochrome P450 enzymes. Conversions above 95% were attained for all the substrates. The key step is the formation of epoxides, which undergo different reaction pathways depending on factors, such as the position of the methyl group and the reaction and work-up conditions used.
