pH-responsive allochroic nanoparticles for the multicolor detection of breast cancer biomarkers

Estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor-2 (HER2) are the three crucial biomarkers for the clinical diagnosis of breast cancer. Sensitive and precise detection of ER, PR, and HER2 is of great significance for the diagnosis of breast cancer. Herein, through a simple solvent-induced self-assembly process, the self-carried allochroic nanoparticles were prepared by using some hydrophobic pH indicator molecules for allochroic NPs-linked immunosorbent assay (named ALISA) of ER, PR, and HER2, respectively. Meanwhile, the introduction of bovine serum albumin (BSA) and antibody (Ab) enhanced the dispersity of the self-assembled nanoparticles as signal tags. Since the ultra-high loading and high-efficiency release of pH indicators, the ALISA exhibitssatisfactory selectivity and sensitivity, which demonstrated the great potential in the early diagnosis and postoperative monitoring of breast cancers. Furthermore, the smartphone was introduced to combine with the ALISA for point-of-care testing, indicating the high feasibility in practical applications.

Superomniphobic Papers for On-Paper pH Sensors

Paper-based superomniphobic surfaces are of great interest because paper is flexible, inexpensive, lightweight, breathable, and recyclable. Prior reports on paper-based superomniphobic surfaces have failed to demonstrate high mobility with low surface tension liquids. In order to overcome this issue, in this work, superomniphobic papers are developed through growth of nanofilaments on inherent microfibers of papers without noticeably altering their microscale features (i.e., diameter and distance of the microfibers). These superomniphobic papers display very low roll-off angle, indicative of ultra-high droplet mobility, even with low surface tension liquids. Here, a facile method is also developed to control the motion and adhesion of the droplets on the superomniphobic paper. Utilizing such liquid mobility in a controlled manner on these superomniphobic papers, a simple on-paper pH sensor is fabricated. It is anticipated that this on-paper, simple, and rapid detection methodology can also be extended to the colorimetric sensing of protein and chemical assays. Further, these superomniphobic papers have potential applications in water?oil separation and enhanced weight-bearing capacity.

Metal-Polydopamine Framework: An Innovative Signal-Generation Tag for Colorimetric Immunoassay

In this work, an innovative enzyme-free colorimetric immunoassay was proposed for the sensitive detection of alpha-fetoprotein (AFP) by introducing thymolphthalein-modified metal-polydopamine framework (MPDA@TP) for the signal generation and amplification. Using zeolitic imidazolate framework (ZIF-67) as the template, the hollow-structured metal-polydopamine framework (MPDA) with high surface recovery and abundant groups was synthesized and functionalized with thymolphthalein (TP) molecules via typical pi-stacking reaction. In the presence of target AFP, an MPDA@TP-linked immunosorbent assay (MLISA) was implemented on the capture antibody-modified microplate by using detection antibody-labeled MPDA@TP as the secondary antibody. Upon alkaline solution introduction, the coated hydrophobic TP on the MPDA was deprotonated into hydrophilic TP2- ion and dissolved in the solution, thereby resulting in the color change of the solution from nearly colorless to deep blue, and the increasing absorbance of the solution at 595 nm. Importantly, the MPDA@TP-based immunoassay could exhibit high sensitivity for the quantitative detection of target AFP on the basis of the absorbance within a linear range of 10-1000 pg mL-1 at a low detection limit of 2.3 pg mL-1. Furthermore, this system was validated preliminarily to screen human serum specimens with well-matched results for the referenced AFP ELISA kit. Taking advantage of simplicity, enzyme-free, convenience, and sensitivity, MPDA@TP-linked immunosorbent assay has the potential for the application in scientific research and clinical diagnosis.

Voltammetric Determination of Quercetin and Rutin on Their Simultaneous Presence on an Electrode Modified with Polythymolphthalein

Abstract: A voltammetric method is developed for the determination of quercetin and rutin in the simultaneous presence on a glassy carbon electrode modified with carbon nanofibers and polythymolphthalein. The conditions of the potentiodynamic electrolysis of thymolphthalein providing the maximum difference in the oxidation potentials of quercetin and rutin equal to 136 mV are found. Thymolphthalein electropolymerization should be carried out in a 100 muM solution by the 10-fold cycling of the potential from 0 to 1.7 V at a sweep rate of 75 mV/s in phosphate buffer solution with pH 7.0. The electrode modified with polythymolphthalein is characterized by scanning electron microscopy, cyclic voltammetry, and electrochemical impedance. It is found that the oxidation of quercetin proceeds irreversibly, and that of rutin, quasi-reversibly with the participation of two electrons and two protons and is controlled by the adsorption of analytes on the electrode surface. The analytical range is 0.025?1.00 muM for both analytes, and the limits of detection are 7.3 nM for quercetin and 4.7 nM for rutin. The selectivity of the electrode response to quercetin and rutin in the presence of inorganic ions, saccharides, and ascorbic and p-coumaric acids is shown. The developed method is tested on real samples (linden Tilia L. flowers and onions).

An automatic system for acidity determination based on sequential injection titration and the monosegmented flow approach

An automatic sequential injection system, combining monosegmented flow analysis, sequential injection analysis and sequential injection titration is proposed for acidity determination. The system enables controllable sample dilution and generation of standards of required concentration in a monosegmented sequential injection manner, sequential injection titration of the prepared solutions, data collecting, and handling. It has been tested on spectrophotometric determination of acetic, citric and phosphoric acids with sodium hydroxide used as a titrant and phenolphthalein or thymolphthalein (in the case of phosphoric acid determination) as indicators. Accuracy better than 4.4% (RE) and repeatability better than 2.9% (RSD) have been obtained. It has been applied to the determination of total acidity in vinegars and various soft drinks. The system provides low sample (less than 0.3 mL) consumption. On average, analysis of a sample takes several minutes.

Adsorption properties of acid dye on amine-terminated dendritic polymer

A series of amine-terminated dendritic polymers (ADPs) were synthesized by stepwise substitution reaction and were used as an adsorbent for removal of acid black dye NT from aqueous solution. Comparison experiments showed that the ADP-3 has good adsorption effect. Then, the effects of various parameters such as initial acid black dye NT concentration (50-300mg/L) and solution pH(1.5- 6.5) were investigated. The point of zero charge (PZC) of ADP-3 has been determined. Specific surface area and average adsorption pore size of the ADP-3 were 21.378m2/g and 3.833nm, respectively. Field scanning electron microscopy (FESEM) indicated that the adsorption material has porous structure, which provided favorable conditions for adsorption process. The experimental data were analyzed by Langmuir and Freundlich models of adsorption. Equilibrium data were fitted well with the Freundlich model. Thermodynamics parameters, DeltaG, DeltaH and DeltaS indicate that the adsorption of acid black dye NT onto ADPs were spontaneous and endothermic process in nature. Pseudo-first-order and pseudo-second-order kinetic models were used to fit the adsorption kinetic data. The adsorption kinetics was found to follow the pseudo-first-order kinetic model. Amino blocking indicated that the amino content plays an important role in the adsorption process. The adsorbent was also applied to the treatment of leather dye effluent, the results showed that ADP-3 exhibit a prominent adsorption property on leather dye effluent.

A specific fluorescent probe for zinc ion based on thymolphthalein and it’s application in living cells

Many diseases are connected with chemical substances, zinc ion is most important ion in chemical substances, because zinc ion (Zn2+) has been shown extensively involved in various physiological and pathological processes. In this work, thymolphthalein with good biocompatibility was introduced, the probe was constructed with interweaved benzimidazole and phenol from into the fluorophore system, hydroxyl and amino creating a multipoint binding pocket, which zinc can coordinate in a cooperative breaking Excited-State Intramolecular Proton Transfer, ESIPT. The fluorescence of this probe adding Zn2+ showed an obvious ?turn on? in a quick response within 5 s. In addition, the probe had a good selectivity for zinc ion over other analytes. Furthermore, we also studied the fluorescence imaging of probe in the presence of Zn2+ in living A549 cells.

Solution properties of dissymmetric sulfonate-type anionic gemini surfactants

Dissymmetric and symmetric anionic gemini surfactants, N-alkyl-N?-alkyl-N,N?dipropanesulfonylethylenediamine (CmCnSul, where m and n represent alkyl chain lengths of m-n = 4-16, 6-14, 8-12, 10-10, and 12-12), were synthesized by two- or three-step reactions. Their physicochemical properties were characterized by equilibrium surface tension measurements, steady-state fluorescence spectroscopy of pyrene, and dynamic light scattering. The critical micelle concentration (CMC) of the dissymmetric surfactants C4C16Sul, C6C14Sul, and C8C12Sul was slightly lower than that of the symmetric surfactant C10C10Sul. The occupied area per molecule (A) of C8C12Sul was smaller than that of C10C10Sul, indicating that C8C12Sul has a high surface activity. However, the increase in the degree of dissymmetry from C8C12Sul to C6C14Sul and then to C4C16Sul resulted in high surface tension and large A. Based on the surface tension, the standard free energies of micellization (?Gmic) and adsorption (?Gads), the efficiency of surface adsorption (pC20), and the effectiveness of surface adsorption (CMC/C20) were obtained. These parameters suggested that C8C12Sul formed micelles more readily than the other surfactants. The properties determined from the surface tension indicated that C8C12Sul?s ability is intermediate between those of C10C10Sul and C12C12Sul. The pyrene fluorescence and dynamic light scattering results revealed that the micelle size depends on the longer of the two alkyl chains in dissymmetric surfactants.

Spectrophotometric determination of nalidixic and pipemidic acids in a flow injection assembly with a solid-phase reactor as a highly stable reagent source

The flow-injection spectrophotometric determination of nalidixic (I) and pipemidic (II) acids were carried out by reaction with Fe(III) ions entrapped in a polymeric material and filling a packed-bed reactor; the released Fe(III) complexed with the quinolone is monitored at 426 nm. The method worked with a high reproducibility, even when using different reactors, days and solutions. Selectivity of the procedure was tested with different foreign compounds found in human urine as well as in pharmaceutical formulations containing both quinolones; no relevant interferences were observed. The calibration graph for (I) was linear over the range 1-250 mugml-1 with a relative standard deviation of 0.7% at 100 mugml-1 (n = 43). The calculated sample throughput was of 26 h-1. The LOD was (s/n = 3) 0.1 mugml-1. The calibration graph for (II) was linear over the range 10-500 mugml-1 with a relative standard deviation of 1.1% at 150 mugml-1 (n = 40). The calculated sample throughput was of 25 h-1 and the LOD was (s/n = 3) 1 mugml-1. The method was applied to determine the (I) in pharmaceutical formulations and of (II) in pharmaceutical formulations and human urine.
