Aiming the faster development from bench to bedside of new potential tracers, multimodal tracers for positron emission tomography (PET) and optical imaging (OI) have emerged as a very promising tool. Indeed, they combine the simplicity of use of optical techniques for in vitro/in vivo pre-clinical studies with the various clinical possibilities offered by PET imaging using their radioactive versions. In this context, the preparation of new tags detectable by fluorescence imaging and potentially suitable for PET imaging after a last-step C-11-labeling of the corresponding precursor has been investigated. Various designs and syntheses were explored by linking o-iodobenzyl alcohols and tetramethyl-BODIPY moieties together. Synthetic polymer-based materials are ubiquitous in aquatic environments, where weathering processes lead to their progressive fragmentation and the leaching of additive chemicals. The current study assessed the chemical content of freshwater and marine leachates produced from car tire rubber (CTR), polypropylene (PP), polyethylene terephthalate (PET), polystyrene (PS) and polyvinyl chloride (PVC) microplastics, and their adverse effects on the microalgae Raphidocelis subcapitata (freshwater) and Skeletonema costatum (marine) and the Mediterranean mussel Mytilus galloprovincialis. A combination of non-target and target chemical analysis revealed a number of organic and metal compounds in the leachates, including representing plasticizers, antioxidants, antimicrobials, lubricants, and vulcanizers. CTR and PVC materials and their corresponding leachates had the highest content of tentatively identified organic additives, while PET had the lowest. The metal content varied both between polymer leachates and between freshwater and seawater. Notable additives identified in high concentrations were benzothiazole (CTR), phthalide (PVC), acetophenone (PP), cobalt (CTR, PET), zinc (CTR, PVC), lead (PP) and antimony (PET). All leachates, except PET, inhibited algal growth with EC50 values ranging from 0.5% (CTR) and 64% (PP) of the total leachate concentration. Leachates also affected mussel endpoints, including the lysosomal membrane stability and early stages endpoints as gamete fertilization, embryonic development and larvae motility and survival. Embryonic development was the most sensitive parameter in mussels, with EC50 values ranging from 0.8% (CTR) to 65% (PET) of the total leachate. The lowest impacts were induced on D-shell larvae survival, reflecting their ability to down-regulate motility and filtration in the presence of chemical stressors. This study provides evidence of the relationship between chemical composition and toxicity of plastic/rubber leachates. Consistent with increasing contamination by organic and inorganic additives, the leachates ranged from slightly to highly toxic to mussels and algae, highlighting the need for a better understanding of the overall impact of plastic-associated chemicals on aquatic ecosystems. We report here the construction of two metal-organic frameworks (MOFs), Zr-6-Cu/Fe-1 and Zr-6–Cu/Fe-2, by integrating earth-abundant cuprous photosensitizers (Cu-PSs) and Fe catalysts for photocatalytic aerobic oxidation. Site isolation and pore confinement stabilize both Cu-PSs and Fe catalysts, while the proximity between active centers facilitates electron and mass transfer. Upon visible light irradiation and using O-2 as the only oxidant, Zr-6-Cu/Fe-1 and Zr-6-Cu/ Fe-2 efficiently oxidize alcohols and benzylic compounds to afford corresponding carbonyl products with broad substrate scopes, high turnover numbers of up to 500 with a 9.4-fold enhancement over homogeneous analogues, and excellent recyclability in four consecutive runs. Control experiments, spectroscopic evidence, and computational studies revealed the photooxidation mechanism: oxidative quenching of [Cu-PS]* by O-2 affords [Cu-II-PS], which efficiently oxidizes Fe-III-OH to generate a hydroxyl radical for substrate oxidation. This work highlights the potential of MOFs in promoting earth-abundant metal-based photocatalysis.

Background: Dipeptidyl peptidase-4 (DPP-4) inhibitors have emerged as anti-hyperglycemic agents that improve glycemic control in type 2 diabetic patients, either as monotherapy or in combination with other antidiabetic drugs. Methods: A novel series of dihydropyrimidine phthalimide hybrids was synthesized and evaluated for their in vitro and in vivo DPP-4 inhibition activity and selectivity using alogliptin as reference. Oral glucose tolerance test was assessed in type 2 diabetic rats after chronic treatment with the synthesized hybrids +/- metformin. Cytotoxicity and antioxidant assays were performed. Additionally, molecular docking study with DPP-4 and structure activity relationship of the novel hybrids were also studied. Results: Among the synthesized hybrids, 10g, 10i, 10e, 10d and 10b had stronger in vitro DPP-4 inhibitory activity than alogliptin. Moreover, an in vivo DPP-4 inhibition assay revealed that 10g and 10i have the strongest and the most extended blood DPP-4 inhibitory activity compared to alogliptin. In type 2 diabetic rats, hybrids 10g, 10i and 10e exhibited better glycemic control than alogliptin, an effect that further supported by metformin combination. Finally, 10j, 10e, 10h and 10d had the highest radical scavenging activity in DPPH assay. Conclusions: Hybrids 10g, 10i and 10e are potent DPP-4 inhibitors which may be beneficial for T2DM treatment.

A highly diastereoselective and enantioselective Michael addition/desymmetrization reaction of maleimides with prochiral 3- substituted phthalides catalyzed by quinine-derived bifunctional thiourea was realized. Air-stable iron carbonyl compounds bearing cyclo-pentadienone ligands with varying substitution were explored ,as catalysts in dehydrogenative diol lactonization reactions using acetone as both the solvent and hydrogen acceptor. Two catalysts with trimethylsilyl groups in the 2- and 5-positions, [2,S-(SiMe3)(2)-3,4-(CH2)(4)(eta(4)-C4C=O))Fe(CO)(3) (1) and [2,5-(SiMe3)(2)-3,4(CH2)(3)(eta(4)-C4C=O)]Fe(CO)(3) (2), were found to be the most active, with 2 being the most selective in the lactonization of diols containing both primary and secondary alcohols. Lactones containing five-, six-, and seven-membered rings .were successfully synthesized, and no over-oxidations to carboxylic acids were detected. The lactonization of unsymmetrical diols containing two primary alcohols occurred with catalyst 1, but selectivity was low based on alcohol electronics and modest based on alcohol sterics. Evidence for a transfer dehydrogenation mechanism was found, and insight into the origin of selectivity in the lactonization of 1 degrees/2 degrees diols was obtained. Additionally, spectroscopic evidence for a trimethylamine-ligated iron species formed in solution during the reaction was discovered.

Antagonists for the ATP-gated ion channel receptor P2X1 have potential as antithrombotics and for treating hyperactive bladder and inflammation. In this study, salicylanilide derivatives were synthesized based on a screening hit. P2X1 antagonistic potency was assessed in 1321N1 astrocytoma cells stably transfected with the human P2X1 receptor by measuring inhibition of the ATP-induced calcium influx. Structure-activity relationships were analyzed, and selectivity versus other P2X receptor subtypes was assessed. The most potent compounds, N-[3,5-bis(trifluoromethyl)phenyl]-5-chloro-2-hydroxybenzamide (1, IC50 0.0192 mu M) and N-[3,5-bis(trifluoromethyl)phenyl]-4-chloro-2-hydroxybenzamide (14, IC50 0.0231 mu M), displayed >500-fold selectivity versus P2X2 and P2X3, and 10-fold selectivity versus P2X4 and P2X7 receptors, and inhibited collagen-induced platelet aggregation. They behaved as negative allosteric modulators, and molecular modeling studies suggested an extracellular binding site. Besides selective P2X1 antagonists, compounds with ancillary P2X4 and/or P2X7 receptor inhibition were discovered. These compounds represent the first potent, non-acidic, allosteric P2X1 receptor antagonists reported to date.

For the metal-catalyzed asymmetric hydrogenation of alpha-substituted ketones, cis reductive products are generally obtained due to steric hindrance of substituents. Herein, an unprecedented trans reductive products were observed in palladium-catalyzed hydrogenative desymmetrization of cyclic and acyclic 1,3-diketones, providing the chiral trans beta-hydroxy ketones with two adjacent stereocenters including one alpha-tertiary or quaternary stereocenter with high enantioselectivity and diastereoselectivity. Mechanistic studies and DFT calculations suggested that the rarely observed diastereoselectivity reversal is ascribed to the charge-charge interaction between the palladium and aromatic ring of the substrate, which could not only result in the reversal of the diastereoselectivity, but also improve the reactivity.
