A highly diastereoselective and enantioselective Michael addition/desymmetrization reaction of maleimides with prochiral 3- substituted phthalides catalyzed by quinine-derived bifunctional thiourea was realized. A broad range of the 3,3-disubstituted phthalides bearing vicinal quaternary-tertiary stereogenic centers were synthesized in moderate to good yields (up to 96%) with high diastereoselectivities (up to >19:1 dr) and enantioselectivities (up to 96:4 er).

We report here the construction of two metal-organic frameworks (MOFs), Zr-6-Cu/Fe-1 and Zr-6–Cu/Fe-2, by integrating earth-abundant cuprous photosensitizers (Cu-PSs) and Fe catalysts for photocatalytic aerobic oxidation. Site isolation and pore confinement stabilize both Cu-PSs and Fe catalysts, while the proximity between active centers facilitates electron and mass transfer. Upon visible light irradiation and using O-2 as the only oxidant, Zr-6-Cu/Fe-1 and Zr-6-Cu/ Fe-2 efficiently oxidize alcohols and benzylic compounds to afford corresponding carbonyl products with broad substrate scopes, high turnover numbers of up to 500 with a 9.4-fold enhancement over homogeneous analogues, and excellent recyclability in four consecutive runs. Control experiments, spectroscopic evidence, and computational studies revealed the photooxidation mechanism: oxidative quenching of [Cu-PS]* by O-2 affords [Cu-II-PS], which efficiently oxidizes Fe-III-OH to generate a hydroxyl radical for substrate oxidation. This work highlights the potential of MOFs in promoting earth-abundant metal-based photocatalysis.

5-Amino-1-phenyl-1H-pyrazole-4-carbohydraizde(1) was used as a key to synthesize new imidazopyrazol-5(1H)-one 5, pyrazolopyrimidines 7 and 9. Furthermore, imidazopyrazoles 13, 15, pyrazolopyridine 16 and pyrazolopyrazolopyridopyrimidine 17 were prepared from pyrazoles 10 and 11. The structure of the new compounds was confirmed by spectral data as well as elemental analysis. Some compounds were selected by the national cancer institute NCI (USA) for anticancer activity against different human cancer cell lines. The results indicated that compound 12 exhibited strong anticancer activity toward Renal Cancer UO-31(GI, 42.81%), while compound 3 was strong active against Breast Cancer MCF7 (GI, 49.88%) and moderate against T-47D (GI, 38.15%) cell lines. However, compound 17 showed strong activity against Leukemia CCRF-CEM (GI, 41.37%) and SR (GI, 44.95%), whereas, compound 9 showed weak activity toward all tested cancer cell lines.

Utilizing a chiral bicyclic imidazole organocatalyst and adopting a continuous injection process, an alternative route has been developed for the efficient synthesis of chiral phthalidyl ester prodrugs via dynamic kinetic resolution of 3-hydroxyphthalides through enantioselective acylation (up to 99 % ee). The computational studies suggest a general base catalytic mechanism differing from the widely accepted nucleophilic catalytic mechanism. The structure analysis of the key transition states shows that the CH-pi interactions and not the previously considered cation/pi-pi interactions between the catalyst and substrate is the dominant factor giving rise to the observed stereocontrol.

The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and carbonucleophiles afforded poly(arylene alkenylene)s with moderate molecular weight in good yield. The reaction involves Mizoroki-Heck coupling, olefin migration via chain walking, and addition of the carbonucleophile to the resulting pi-allylpalladium species. The polymerization with a slight excess of nucleophile with respect to diiodoarene also proceeded to give the polymer without significant decrease in molecular weight in spite of the nonstoichiometric mixture of the monomers. The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and diimide also proceeded. The base used in the reaction is critical for yield and molecular weight of the product. The reaction using NaHCO3 afforded the product with low solubility, which can be explained by the high molecular weight of the polymer and/or the strong interaction of the electron donating dimethoxyphenylene groups and electron accepting diimide groups in the polymer. Phthalate derivatives with low estrogenic activity, high infrared spectrum signals, high Raman characteristic vibration spectrum, high fluorescence intensity, and high ultraviolet sensitivity were selected as precursors from our previous studies, so that the changes in their toxicity and estrogenic activity during biological metabolism, ozone oxidation, photocatalytic degradation, photodegradation, and microbial degradation could be studied.The transformation pathways of these derivatives were simulated, and the reaction energy barriers were calculated. To determine the potential environmental risks of these phthalate derivatives, the pharmacophore models of biotoxicity and estrogen activity of phthalates were used to predict the biotoxicity and estrogen activity of the transformed products. The results showed an increase in the biotoxicity and estrogen activity of the biometabolites, ozonation products, photocatalytic degradation products, and microbial degradation products; the only products that did not follow this trend were the photodegradation products. Notably, the pathways that produced more potentially toxic compounds were the less favorable paths. Our results indicate that the transformation products of the designed environmentally friendly phthalate derivatives potentially pose environmental risks. To avoid such risks, the environmental transformation pathway of these derivatives should be simulated to screen for environmentally friendly phthalate molecules. Environ Toxicol Chem 2020;00:1-11. A research program to discover solubilizing prodrugs of the HCV NS5A inhibitor pibrentasvir (PIB) identified phosphomethyl analog 2 and trimethyl-lock (TML) prodrug 9. The prodrug moiety is attached to a benzimidazole nitrogen atom via an oxymethyl linkage to allow for rapid and complete release of the drug for absorption following phosphate removal by intestinal alkaline phosphatase. These prodrugs have good hydrolytic stability properties and improved solubility compared to PIB, both in aqueous buffer (pH 7) and FESSIF (pH 5). TML prodrug 9 provided superior in vivo performance, delivering high plasma concentrations of PIB in PK studies conducted in mice, dogs, and monkeys. The improved dissolution properties of these phosphate prodrugs provide them the potential to simplify drug dosage forms for PIB-containing HCV therapy.

In this work, we synthesized thulium (Tm3+) modified TiO2 nanoparticles by a facile sol-gel route. We show that Tm3+-modified TiO2 exhibit superior photocatalytic activity than pure TiO2 towards gas-phase degradation of acetaldehyde, o-xylene, and their mixture. The photocatalytic activity enhances with the increasing Tm content from 0.1 to 0.5 mol%. The 0.5 mol%Tm-TiO2 sample demonstrated the highest photodegradation efficiency (> 90.0%) for acetaldehyde and o-xylene. According to the systematic characterization such as photoluminescence (PL), photocurrent, up-conversion spectra, UV-Vis diffuse reflectance spectroscopy and electron spin resonance (ESR) analysis, the enhanced photocatalytic performance can be attributed to introducing the new Tm-4f states between the original band gap of TiO2, which led to the increase of efficiency of light-induced electron-hole separation and the increase of hydrophilicity which resulted in the increased adsorption capacity towards acetaldehyde and o-xylene. Gas Chromatography-Mass Spectrometer (GC-MS) analysis suggested that o-xylene was first oxidized to o-benzaldehyde, which was eventually converted to butanol before being mineralized into CO2. However, new intermediates including phthalide, o-methyl acetophenone, naphthalene and 5-Methyl-4-octanone were detected in the mixture.
