When the faded documents surfaces are exposed to the thermal effect there is no change in all the faded documents. Adaptive antibiotic resistance is a newly described phenomenon by which Acinetobacter baumannii induces efflux pump activity in response to host-associated environmental cues that may, in part, account for antibiotic treatment failures against clinically defined susceptible strains. To that end, during adaptation to growth in human serum, the organism induces approximately 22 putative efflux-associated genes and displays efflux-mediated minocycline tolerance at antibiotic concentrations corresponding to patient serum levels. Here, we show that in addition to minocycline, growth in human serum elicits A. baumannii efflux-mediated tolerance to the antibiotics ciprofloxacin, meropenem, tetracycline, and tigecycline. Moreover, using a whole-cell highthroughput screen and secondary assays, we identified novel serum-associated antibiotic efflux inhibitors that potentiated the activities of antibiotics toward serum-grown A. baumannii. Two compounds, Acinetobacter baumannii efflux pump inhibitor 1 (ABEPI1) [(E)-4-((4-chlorobenzylidene)amino)benezenesulfonamide] and ABEPI2 [N-tert-butyl-2-(1-tert-butyltetrazol-5-yl) sulfanylacetamide], were shown to lead to minocycline accumulation within A. baumannii during serum growth and inhibit the efflux potential of the organism. While both compounds also inhibited the antibiotic efflux properties of the bacterial pathogen Pseudomonas aeruginosa, they did not display significant cytotoxicity toward human cells or mammalian Ca2+ channel inhibitory effects, suggesting that ABEPI1 and ABEPI2 represent promising structural scaffolds for the development of new classes of bacterial antibiotic efflux pump inhibitors that can be used to potentiate the activities of current and future antibiotics for the therapeutic intervention of Gram-negative bacterial infections.

An enzyme-free titer plate-based colorimetric assay utilizing functionalized mesoporous silica nanoparticles (MSNs) entrapping pH-indicator molecules has been developed. Pores in the silica nanoparticles were functionalized with phenyltrimethyloxysilane so that pH indicator molecules (thymolphthalein or TP in the present case) can be tightly entrapped through pi-pi conjugation. To detect prostate specific antigen (PSA), the TP-containing MSNs were coated with polyethylenimine (PEI), which favors the attachment of the negatively charged secondary anti-PSA antibody. The entrapped thymolphthalein molecules can be readily released from the pores with a simple addition of alkaline solution. The resultant bifunctional MSNs were used for signal-amplified detection of PSA captured by the primary antibody preimmobilized in the wells of a plate. Our method possesses a wide dynamic range (0.5 to 8000 pg/mL) wherein the adsorption of the bifunctional MSNs obeys a modified Langmuir isotherm. A detection limit (LOD) down to as low as 0.36 pg/mL can be attained. Owing to the size uniformity of the MSNs and the obviation of enzyme molecules employed in the enzyme-linked immunosorbent assay (ELISA), excellent reproducibility (RSD = 1.12%) was achieved. The selective detection of PSA in human serum samples demonstrates the amenability of our method to detect important biomarkers in complex biological samples, whereas the performance of the assay in a titer plate ensures high throughput and obviates the use of expensive instruments. Both of these features are prerequisites for clinical settings wherein a great number of samples need to be analyzed in a timely fashion.

Mixtures of benzoic acid with various organic compounds are subjected to plastic deformation in Bridgman anvils under a pressure of 1 GPa at room temperature. This study aimed to investigate the influence of a three-dimensional cell culture model and bioactive glass (BG) particles on the expression of osteoblastic phenotypes in rat calvaria osteogenic cells culture. Cells were seeded on two-dimensional (2D) and threedimensional (3D) collagen with BG particles for up to 14 days. Cell viability and alkaline phosphatase (ALP) activity was performed. Cell morphology and immunolabeling of noncollagenous bone matrix proteins were assessed by epifluorescence and confocal microscopy. The expressions of osteogenic markers were analyzed using RT-PCR. Mineralized bone-like nodule formation was visualized by microscopy and calcium content was assessed quantitatively by alizarin red assay. Experimental cultures produced a growing cell viability rate up to 14 days. Although ALP activity at 7 days was higher on BG cultures, cells on 3D and 3D+BG had an activity decrease of ALP at 14 days. Three-dimensional conditions favored the immunolabeling for OPN and BSP and the expression of ALP and COL I mRNAs. BG particles influenced positively the OC and OPN mRNAs expression and calcified nodule formation in vitro. The results indicated that the 3D cultures and BG particles contribute to the expression of osteoblastic phenotype and to differentiated and mineralized matrix formation.

Essential oils (EO) are complex mixtures of biosynthesized chemicals basically by plants, which provide them their characteristic aroma. Many have biologically recognized activities as antioxidants and anti-inflammatory among others, and many of them are employed as cosmetic actives. Very often, these properties are not fully exploited because of their high volatility and tendency to oxidize, so it is necessary to attach them to a conveyor to provide them adequate stabilization and lifetime. One of the best alternatives to carry out this is microencapsulation, for which natural biopolymers can be used, such as the starches. Therefore, it used EO of thyme, cinnamon, and clove, which were obtained by conventional and assisted hydrodistillation by microwave radiation from the plant material. The chemical composition was evaluated by gas chromatography/mass spectrometry (GC/ MS). The radical scavenging ability was determined by antiradical activity techniques including DPPH (2,2-diphenyl-1-picrylhydrazyl) and ABTS (2,2?-azino-bis-(3-ethylthiazoline-benzenesulfonic acid-6)), and the ORAC method was used for determining the antioxidant capacity. Also, starches of yam (D. rotundata), sweet potato (I. batatas), corn (Z. mays), and cassava (M. esculenta) were used, which were subjected to hydrolysis and lipophilization processes using dodecenyl succinic anhydride (DDSA); this chemical process achieves a significant increase in emulsifier capacity (surfactant) compared to its native state; that of cassava was the most promising starch which was used as an EO microencapsulating agent. Finally, from the EO microcapsules, an emulgel-type cosmetic was designed which maintained its antioxidant activity. The results of this work contribute to the development of stable and functional cosmetic formulations of essential oils, emphasizing that the extraction of EO by assisted microwave radiation hydrodistillation is considered a fast, efficient, green, and relatively economical method compared to conventional hydrodistillation.

We aimed to evaluate the in vitro osteogenic and osteoinductive potentials of BioS-2P and its ability to promote in vivo bone repair. To investigate osteogenic potential, UMR-106 osteoblastic cells were cultured on BioS-2P and Bioglass 45S5 discs in osteogenic medium. The osteoinductive potential was evaluated using mesenchymal stem cells (MSCs) cultured on BioS-2P, Bioglass 45S5 and polystyrene in non-osteogenic medium. Rat bone calvarial defects were implanted with BioS-2P scaffolds alone or seeded with MSCs. UMR-106 proliferation was similar for both materials, while alkaline phosphatase (ALP) activity and mineralization were higher for BioS-2P. Bone sialoprotein (BSP), RUNX2 and osteopontin (OPN) gene expression and BSP, OPN, ALP and RUNX2 protein expression were higher on BioS-2P. For MSCs, ALP activity was higher on Bioglass 45S5 than on BioS-2P and was lower on polystyrene. All genes were highly expressed on bioactive glasses compared to polystyrene. BioS-2P scaffolds promoted in vivo bone formation without differences in the morphometric parameters at 4, 8 and 12 weeks. After 8 weeks, the combination of BioS-2P with MSCs did not increase the quantity of new bone compared to the BioS-2P alone. To stimulate osteoblast activity, drive MSC differentiation and promote bone formation, BioS-2P is a good choice as a scaffold for bone tissue engineering.

This study aimed to investigate the toxicity of BTEX (benzene, toluene, xylene and ethylbenzene) on a microcosm scale using the MS-3 multispecies system. The multispecies system (MS-3) was employed to investigate the BTEX diluted in water and ethanol. The columns for the test were prepared in PVC tubes, open at the top and the bottom was covered by a mesh. In each system were added soil macro-decomposers (earthworm and woodlice) and plant species (mustard and lettuce). The soil used in the column was dried beforehand and sieved. It was placed in MS-3 system up to 20 cm height of soil and approximately 6 kg of soil. During the 14 days of experiment, five applications of BTEX were performed and, at the end of the period, the growth and survival of organisms and enzymatic activity in the soil were analyzed. After the exposure time, it observed the decrease in the weight of the woodlice and a reduction in lettuce and mustard seed germination in BTEX water system, while there was no statistically significant difference between the BTEX + ethanol system and their respective control. There were no changes of mortality in macro-invertebrates, length of seedlings or alterations of the soil microbial activity. With our data, we conclude that the implementation of this model to evaluate the quality of the soil constitutes a promising alternative in ecotoxicological investigations and that the BTEX can affect soil organisms in different ways.
