The isobenzofuran-1(3H)-ones (phthalides) exhibit various biological activities, including antioxidant activity on reactive oxygen species (ROS). An excess of ROS that cannot be naturally contained by cellular enzymatic systems is called redox imbalance, which damage cell membranes, proteins, and DNA, thereby possibly triggering neuronal death in several neurodegenerative diseases. Considering our ongoing efforts to find useful compounds to control redox imbalance, herein we evaluated the antioxidant activity of two phtalides (compounds 3 and 4), using primary cultures of hippocampal neurons. Spectrophotometric assays showed that compound 3 significantly reduced (p <= 0.05) ROS levels and lipid peroxidation compared to the control treatment, while compound 4 was unable at any of the tested concentrations. Despite their structural similarity, these compounds behave differently in the intracellular environment, which was reliably corroborated by the determination of oxidation potentials via cyclic voltammetry. It was demonstrated that compound 3 presents a lower oxidation potential. The combination of the mentioned methods allowed us to find a strong correlation between the chemical structure of compounds and their biological effects. Taking together, the results indicate that compound 3 presents desirable characteristics to act as a candidate pharmacological agent for use in the prevention and treatment of neurodegenerative diseases. Plasma catalysis has been utilized in many environmental applications for removal of various hydrocarbons including tars. The aim of this work was to study the tars removal process by atmospheric pressure DBD non-thermal plasma generated in combination with packing materials of various composition and catalytic activity (TiO2, Pt/gamma Al2O3, BaTiO3, gamma Al2O3, ZrO2, glass beads), dielectric constant (5-4000), shape (spherical and cylindrical pellets and beads), size (3-5 mm in diameter, 3-8 mm in length), and specific surface area (37-150 m(2)/g). Naphthalene was chosen as a model tar compound. The experiments were performed at a temperature of 100 degrees C and a naphthalene initial concentration of approx. 3000 ppm, i.e., under conditions that are usually less favorable to achieve high removal efficiencies. For a given specific input energy of 320 J/L, naphthalene removal efficiency followed a sequence: TiO2 > Pt/gamma Al2O3 > ZrO2 > gamma Al2O3 > glass beads > BaTiO3 > plasma only. The efficiency increased with the increasing specific surface area of a given packing material, while its shape and size were also found to be important. By-products of naphthalene decomposition were analyzed by means of FTIR spectrometry and surface of packing materials by SEM analysis.

Kinetics of thermostimulated luminescence (TSL) of poly(diphenylenephthalide) (PDPh) films has been studied using mathematical modelling and DFT approximation. TSL of PDPh-film was considered to be arising from irradiative deactivation of diphenylene (DP) fragment of polymer chain in excited state formed upon recombination of the spatial separated ion-radical pairs generated by photoexcitation. Two parallel monomolecular transformations leading to excited DP have been revealed: the first path is characterized by the activation parameters of the kinetic rate constant k1 which equal to Al ti 105 sec 1 and Eat ti 70 kJ mol(-1). This path may be interpreted in terms of mechanical relaxation theory by segmental polymer mobility as electron transfer (ET) from phthalide (Ph) anion-to DP cation-radical. The second channel is supposed to be consist of two consecutive ET from triarylmethyl anion-radical to phthalide neutral (A2 ti 1 sec 1, Ea2 ti 20 kJ mol(-1)), and then to DP cation-radical (A3 N 104 sec 1, E,3 N 40 kJ mol(-1)) of the polymer chain indicating y- and fl-relaxation, respectively. The revealed mechanistic steps and electron retention efficiency estimated in B97-2/6-311 + G(d,p) approximation for the most probable anion-radicals may point out the key role of domino quantum tunneling in transfer and recombination of charges in PDPh films.

Comprehensive two-dimensional gas chromatography with time-of-flight mass spectrometry (GC x GC-TOF-MS) was combined with conventional mono-dimensional GC-MS to differentiate Croatian virgin olive oils (VOO) according to variety and geographical origin, based on the profile of volatile aroma compounds isolated by HS-SPME. More than 1000 compounds were detected after untargeted profiling and 256 were identified or tentatively identified, providing one of the most detailed profiles of volatile aroma compounds in VOO up to date. Among them, 131 volatile compounds were significantly different across monovarietal VOOs, while 60 were found useful for the discrimination according to geographical origin. Many major lipoxygenase and minor non-lipoxygenase-generated compounds were shown to have discriminating ability with respect to both factors. Multivariate statistical analysis extracted twenty-one volatile markers with the highest discriminant power for varietal differentiation. The approach reported may have practical application in better understanding, defining, managing, and communicating the varietal or geographical typicity of monovarietal VOOs.

The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and carbonucleophiles afforded poly(arylene alkenylene)s with moderate molecular weight in good yield. The reaction involves Mizoroki-Heck coupling, olefin migration via chain walking, and addition of the carbonucleophile to the resulting pi-allylpalladium species. The polymerization with a slight excess of nucleophile with respect to diiodoarene also proceeded to give the polymer without significant decrease in molecular weight in spite of the nonstoichiometric mixture of the monomers. The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and diimide also proceeded. The base used in the reaction is critical for yield and molecular weight of the product. The reaction using NaHCO3 afforded the product with low solubility, which can be explained by the high molecular weight of the polymer and/or the strong interaction of the electron donating dimethoxyphenylene groups and electron accepting diimide groups in the polymer. Photoswitchable molecules have multiple applications in the physical and life sciences because their properties can be modulated with light. Fluxional molecules, which undergo rapid degenerate rearrangements in the electronic ground state, also exhibit switching behavior. The stochastic nature of fluxional switching, however, has hampered its application in the development of functional molecules and materials. Here we combine photoswitching and fluxionality to develop a fluorophore that enables very long (>30 min) time-lapse single-molecule localization microscopy in living cells with minimal phototoxicity and no apparent photobleaching. These long time-lapse experiments allow us to track intracellular organelles with unprecedented spatiotemporal resolution, revealing new information of the three-dimensional compartmentalization of synaptic vesicle trafficking in live human neurons.

Fourteen new compounds including thirteen drimane – phthalide derivatives (fendlerals A – C, fendlerins A – D, fendlerols A – B, fendleric acids A – C, fendlerinine G) and one terphenyl derivative (fendleryl E) along with eight known compounds, fendlerinine A, rickenyls C – D, fendleryls C – D, atromentin, tetramethyl atromentin, and ( +/- )-microsphaerophthalide F, were isolated from the wood fungus Hypoxylon fendleri BCC32408. Compared with the prior work, the results indicated the agitation effect on the production of bioactive drimane – phthalides. The chemical structures were determined based upon spectroscopic analyses and the absolute configurations were verified by comparison of the ECD spectral data with the calculated ECD spectra of the related compounds. Compounds 1-3 exhibited antimicrobial activity against Plasmodium falciparum (IC50 4.15-4.39 mu M), Colletotrichum capsici (MIC 6.25-12.5 mu g/mL), and Bacillus cereus (MIC 1.56-3.13 mu g/mL). All tested compounds displayed broad cytotoxicity against cancerous (MCF-7, KB, and NCI-H187) and non-cancerous (Vero) cells.
