Starting from micromolar chalcone hits, a predictive QSAR model for diaryl propenones was developed, and synthetic analogues with a 100-fold increase in potency were obtained. Two nanomolar hits underwent efficacy validation and eADMET profiling; one compound was shown to increase the survival time in an artificial metastasis model in nude mice. Although the molecular mechanism(s) by which these substances mediate efficacy remain(s) unrevealed, we were able to eliminate the major targets commonly associated with antineoplastic chalcones.

A series of novel 1-((benzofuran-2-yl)methyl)-1H-triazole derivatives has been synthesized and tested in vitro against a panel of five different human tumor cell lines. The results show that the existence of benzotriazole or 1,2,3- triazole ring and substitution of the triazolyl-3-position with a naphthylacyl, 4-bromophenacyl or 4-methylbenzyl group could be crucial for promoting cytotoxic activity. Compounds 18, 19, 20 and 25 were found to have the most potent activities selectively against HL-60, SMMC-7721 and MCF-7 cell lines respectively. In particular, compound 20 was more selective towards HL-60 and A549 cell lines with IC50 values of 0.62 and 1.60 muM.

Optical and chiroptical photoswitches have found potential applications in advanced information technologies. Herein, a pair of chiral triple-stranded helicates [Eu2(o-LRR)3](TOf)6and [Eu2(o-LSS)3](TOf)6, which have chirality, luminescence and light stimuli-responsive properties, have been designed and synthesized. The ligands o-LRR/SSare composed of photochromic diarylethene moieties as spacers and two chiral 2,6-dipicolinic amides as coordination units. Through the comprehensive spectral characteristics in combination with semiempirical geometry optimization using the Sparkle/RM1 model, it was confirmed that (R/S)-1-phenylethanamines decorated at the terminals of ligands successfully controlled the stereoselective self-assembly of the helicates. Upon alternating UV and visible light irradiation, the diarylethene experienced a reversible change between a colorless open-ring state and a pink closed-ring state. This photochromic process caused variations in the electronic structure and ligand conformation, and led to the presence of light-responsive optical (UV and PL) and chiroptical (ECD and CPL) switching properties. The helicate may, therefore, be considered as a completely reversible +/- ECD and on/off CPL switch. Furthermore, on the basis of these light-switchable optical properties, the INHIBIT and IMPLICATION logic gates were fabricated.

Antioxidant properties of 3-hydroxycoumarin derivatives

A series of hydroxylated 3-hydroxycoumarins was synthesised and evaluated for their antioxidant properties. The compounds substituted on the C-7 position were almost as antioxidant as quercetin or vitamin C. The antioxidant properties were related by an EPR study to their abilities to give stable semiquinonic or polyhydroxylated radicals. A series of hydroxylated 3-hydroxycoumarins was synthesised by the reaction of 3-aryl-2-hydroxypropenoic derivatives with boron tribromide. They were evaluated for their ability to scavenge the 2,2-diphenyl-1-picrylhydrazyl radical, the superoxide anion radical, the hydroxyl radical and the peroxynitrite anion and to inhibit copper-induced human LDL peroxidation. The physicochemical results were in accordance to establish the compounds hydroxylated on C-6 and C-7 positions as the most active of the series with antioxidant potencies comparable to those of quercetin and vitamin C. These compounds form o- and p-quinonoid derivatives upon radical scavenging and may serve as new lead compounds for pharmacological investigations.

Synthesis and Cytotoxicity of Octahydroepoxyisoindole-7-carboxylic Acids and Norcantharidin?Amide Hybrids as Norcantharidin Analogues

Octahydroepoxyisoindole analogues of norcantharidin were accessed through a Diels?Alder reaction of an amine-substituted furan with maleic anhydride and subsequent reduction of the bicyclo[2.2.1]heptene olefin. Despite retention of the carboxylate and the ether bridgehead known to impart cytotoxic activity to norcantharidin, none of these analogues displayed notable cytotoxicity against the 11 cell lines examined: HT29 (colon), MCF-7 (breast), A2780 (ovarian), H460 (lung), A431 (skin), Du145 (prostate), BE2-C (neuroblastoma), SJ-G2 and U87 (glioblastoma), MIA (pancreatic), and SMA (spontaneous murine astrocytoma). The incorporation of an amino-substituted system post-synthesis of norcantharidin afforded facile access to 14 acid/amide-substituted norcantharidin analogues. Of these, only four displayed sufficient activity at the initial 25 mum compound screening dose to warrant full evaluation of growth inhibition. Common to these analogues was the presence of a 4-biphenyl moiety, and in particular 3-(2-(furan-2-ylmethyl)-3-(4-biphenylamino)-3-oxopropylcarbamoyl)-7-oxabicyclo[2.2.1]heptane-2-carboxylic acid (13 c) and 3-(2-(pyrrole-2-ylmethyl)-3-(4-biphenylamino)-3-oxopropylcarbamoyl)-7-oxabicyclo[2.2.1]heptane-2-carboxylic acid (24) displayed high levels of cytotoxicity, returning GI50 values of 15 nm (HT29) to 2.9 mum (U87) and 17 nm (SMA) to 2.8 mum (U87), respectively. These are the most cytotoxic norcantharidin analogues reported to date.
