The viscosity properties of acetylated carboxymethyl starch (ACMS) aqueous solution with different DS (degree of substitution) at concentration of 2.0% (w/w) were investigated using Brookfield R/S+ Portable rheometer. All flow curves of ACMS are well fitted to Power law model. Arrhenius model was used to describe temperature sensitivity. Furthermore, the parameters, which influenced the viscosity properties of the solutions, including temperature, DS of carboxymethyl group (DScm), DS of acetyl group (DSac), presence of salt (NaCl) and shear rate were discussed in detail. Specifically, the viscosity-stability, salt-tolerance and shear-resistance of ACMS with DScm (0.76) and DSac (0.34) were compared with that of sodium alginate as thickener in food or non-food applications. The results indicate that the viscosity properties of ACMS (DScm = 0.76, DS ac = 0.34) were similar to that of sodium alginate and ACMS with DScm (0.76) and DSac (0.34) may be considered as a substitute for sodium alginate in certain application.

Objective Fresh-frozen bone allograft (FFBA) is an alternative to autogenous bone (AB) for reconstructing maxillary bone. Despite the promising clinical results, cell responses to FFBA and AB were not evaluated. Thus, our aim was to compare cells harvested from maxillary reconstructed sites with either AB or FFBA in terms of osteoblast differentiation and to evaluate the effect of culturing cells in contact with FFBA. Methods Cells harvested from three patients submitted to bilateral maxillary reconstruction with AB and FFBA were cultured to evaluate: proliferation, alkaline phosphatase activity, extracellular matrix mineralization and gene expression of osteoblastic markers. The effect of FFBA on osteoblast differentiation was studied by culturing cells harvested from AB in contact with FFBA and evaluating the same parameters. Data were compared using either two-way ANOVA followed by Tukey-b test or Student’s t test (p ? 0.05). Results Cell proliferation was higher in cultures from AB grafted sites and extracellular matrix mineralization was higher in cultures derived from FFBA grafted sites. The gene expression of alkaline phosphatase, RUNX2, bone sialoprotein and osteocalcin was higher in cells derived from FFBA compared with cells from AB grafted sites. However, the exposure of cells derived from AB to FFBA particles did not have any remarkable effect on osteoblast differentiation. Conclusions These results indicate the higher osteogenic activity of cells derived from FFBA compared with AB reconstructed sites, offering an explanation at cellular level of why FFBA could be a suitable alternative to AB for reconstructing maxillary bone defects.

An investigation of particle disintegration was carried out using batch anaerobic reactors and a particle-rich substrate from pig manure supernatant. Two types of samples were applied, one high in suspended particles (raw feed) and another low in suspended particle content (centrifuged feed). Both feeds were digested with and without cellulase enzyme addition to obtain a better understanding of particle degradation mechanisms. An automatic methane potential test system (AMPTS) was used to carry out batch reactions at 35 C. The raw feed with high-suspended solids had higher biomethane potential than the centrifuged feed but the conversion rate and methane yield was lower. The addition of cellulase increased biomethane production rates in both high- and low-particle content samples enhancing yield by 54% and 40%, respectively and converting 69% and 87% of feed chemical oxygen demand (COD), respectively. This implies that the feed particles have high contents of cellulose. This is also the case for the smaller particles remaining after centrifugation. Comparisons of anaerobic digestion model no. 1 (ADM1) simulations with experimental data reveal that classifying substrate particles into a fast and a slow degrading fraction with separate disintegration kinetics fit the experimental data better than lumping all particles into one parameter.
