Microtubule-targeting agents that bind at the colchicine-site of tubulin are of particular interest in antitumoral therapy due to their dual mechanism of action as antimitotics and vascular disrupting agents. Cyclohexanediones derivatives have been described as a new family of colchicine-domain binders with an association constant to tubulin similar to that of colchicine. Here, the high-resolution structures of tubulin in complex with cyclohexanediones TUB015 and TUB075 were solved by X-ray crystallography. A detailed analysis of the tubulin-TUB075 interaction by means of computational affinity maps allowed the identification of two additional regions at the binding site that were addressed with the design and synthesis of a new series of cyclohexanediones with a distal 2-substituted benzofurane. These new compounds showed potent antiproliferative activity with IC50 values in the nM range, arrested cell cycle progression at the G2/M phase and induced apoptosis at sub muM concentrations. Moreover, they caused the destruction of a preformed vascular network in vitro and inhibited the migration of endothelial cells at non-toxic concentrations. Finally, these compounds displayed high affinity for tubulin as substantiated by a K b value of 2.87 × 108 M-1 which, to the best of our knowledge, represents the highest binding constant measured to date for a colchicine-domain ligand.

The lighter density (<1.5 g/cm3) fractions produced from two waste coals sampled from the waste coal disposal sites at thermochemical plants situated in South Africa were used as feed materials for liquefaction with tetralin. The liquefaction residues from the lighter density and untreated lighter density fractions were used in pyrolysis experiments. Pyrolysis of the lighter density fractions was carried out in a Fischer Assay oven at 750 and 920 C under an argon atmosphere. Advanced analytical techniques (gas chromatography-mass spectrometry and nuclear magnetic resonance (NMR) spectroscopy) were employed to characterize the pyrolysis products. Also, the lighter density fractions, liquefaction residues, and their chars were examined using conventional and advanced analytical techniques. The pyrolysis char yields of the liquefaction residues ranged between 74% and 76%, and those of the coal float fractions ranged between 67.0 and 71.5%. Gas pyrolysis yields ranged between 16.0% and 20.0% for the residues and between 14.5% and 18.4% for the lighter density fractions, while the pyrolytic water and the tar products of the lighter density fractions were slightly higher than those of the liquefaction-derived residues. The proton NMR analysis of the tars from the residues shows marginally higher amounts of aromatic protons than those of the lighter density fractions. Chars which were generated after pyrolysis of the liquefaction-derived residues show higher porosity values than those from the pyrolysis of the lighter density fractions. The differences in the porosities are attributed to the opening of pores and extraction of some lower molecular mass aliphatic species from the coal matrix during liquefaction. The pyrolysis products distribution and characterization of the products showed that the residues (waste material) generated after tetralin liquefaction of the float fractions from the float-sink experiments of waste coals may be utilized for thermochemical processes (pyrolysis). Volatile components in Dalbergia cochinchinensis Pierre were analysed using a comprehensive two-dimensional gas chromatography with mass spectrometry method featuring a new solid-state modulator. Compared to one-dimensional gas chromatography, the number of detected peaks were significantly increased. A total of 45 major compounds were identified in this study and the forward and reverse match factors of these compounds were both above 800. The results showed that the volatile components in Dalbergia cochinchinensis Pierre were primarily aldehyde?and ketone compounds such as benzaldehyde, cinnamaldehyde, 4-chromanone, 1-(2-hydroxyphenyl)ethanone and acetophenone. In addition, a semi-quantitative analysis was conducted to determine the contents of the detected compounds based on peak area percentage. Moreover, the repeatability of the comprehensive two-dimensional gas chromatography-mass spectrometry analysis in this study was quite satisfactory with relative standard deviations less than 12.7% for intraday and 17.3% for interday measurements.

Heterocyclic aromatic compounds (NSO-HET) have frequently been detected in the environment. Several studies have concluded that NSO-HET pose a threat to organisms in waters, sediments and soils. However, few publications are available assessing the ecotoxicology of NSO-HET. The present study aims to assess the embryo toxicity of heterocycles using Danio rerio. A combination of the Fish Embryo Toxicity Test and analytical quantification should aid to determine the hazard potential. Changes of the total concentrations due to sorption or volatility were quantified by GC/MS. Loss of compounds during the test was observed primarily for volatile or hydrophobic NSO-HET. The LC50 calculated with nominal concentrations underestimates the toxicity by a factor up to 16 (2. h), demonstrating that a chemical analysis for comparing nominal and measured concentrations is essential for such investigations.

A natural fiber reinforced composite, belonging to the class of eco composites, based on ethylene-propylene-terpolymer rubber (EPDM) and wood wastes were obtained by electron beam irradiation at 75, 150, 300, and 600 kGy in atmospheric conditions and at room temperature using a linear accelerator of 5.5 MeV. The sawdust (S), in amounts of 5 and 15 phr, respectively, was used to act as a natural filler for the improvement of physical and chemical characteristics. The crosslinking effects were evaluated through sol-gel analysis, mechanical tests, and Fourier Transform Infrared FTIR spectroscopy comparatively with the classic method with dibenzoyl peroxide (P) applied on the same types of samples at high temperature. Gel fraction exhibits values over 98% but, in the case of P cross-linking, is necessary to add more sawdust (15 phr) to obtain the same results as in the case of electron beam (EB) cross-linking (5 phr/300 kGy). Even if the EB cross-linking and sawdust addition have a reinforcement effect on EPDM rubber, the medium irradiation dose of 300 kGy looks to be a limit to which or from which the properties of the composite are improved or deteriorated. The absorption behavior of the eco-composites was studied through water uptake tests.

Bisphenol A (BPA), an endocrine disrupting compound largely used in plastic and paper industry, ends up in aquatic systems via wastewater treatment plants (WWTPs) among other sources. The identification and quantification of BPA in wastewater (WW) and wastewater sludge (WWS) is of major interest to assess the endocrine activity of treated effluent discharged into the environment. Many treatment technologies, including various pre-treatment methods, such as hydrolysis, Fenton oxidation, peroxidation, ultrasonication and ozonation have been developed in order to degrade BPA in WW and WWS and for the production of WWS based value-added products (VAPs). WWS based VAPs, such as biopesticides, bioherbicides, biofertilizers, bioplastics and enzymes are low cost biological alternatives that can compete with chemicals or other cost intensive biological products in the current markets. However, this field application is disputable due to the presence of these organic compounds which has been discussed with a perspective of simultaneous degradation. The pre-treatment produces an impact on rheology as well as value-addition which has been reviewed in this paper. Various analytical techniques available for the detection of BPA in WW and WWS are also discussed. Presence of heavy metals and possible thermodynamical behavior of the compound in WW and WWS can have major impact on BPA removal, which is also included in the review.
