(Chemical Equation Presented) Herein we find that proazaphosphatrane 1c is a very efficient catalyst for Mukaiyama aldol reactions of aldehydes with trimethylsilyl enolates in THF solvent. Only the activated ketone 2,2,2-trifluoroacetophenone underwent clean aldol product formation with a variety of trimethylsilyl enolates under similar conditions as the aldehydes. The reactions were carried out at room temperature using (1-methoxy-2-methyl-1- propenyloxy)trimethylsilane, whereas the temperature was -15 C in the case of 1-phenyl-1-(trimethylsilyloxy)ethylene. The reaction conditions are mild and operationally simple, and a variety of aryl functional groups, such as nitro, amino, ester, chloro, trifluoromethyl, bromo, iodo, cyano, and fluoro groups, are tolerated. Product yields are generally better than or comparable to those in the literature. 1-Phenyl-1-(trimethylsilyloxy)ethylene, 1-(trimethylsilyloxy) cyclohexene, and 2-(trimethylsilyloxy)furan underwent clean conversion to beta-hydroxy carbonyl compounds under our reaction conditions. In the case of bulky (2,2-dimethyl-1-methylenepropoxy)trimethylsilane, only alpha,beta-unsaturated esters were isolated. Heterocyclic aldehydes, such as pyridine-2-carboxaldehyde, benzofuran-2-carboxaldehyde, benzothiophene-2- carboxaldehyde, and 1-methyl-2-imidazolecarboxaldehyde, gave good yields of Mukaiyama products. An optimized synthesis for the catalyst 1c is also reported herein. 2009 American Chemical Society.

Enantioselective aldol reactions with masked fluoroacetates

Despite the growing importance of organofluorines as pharmaceuticals and agrochemicals, the stereoselective introduction of fluorine into many prominent classes of natural products and chemotherapeutic agents is difficult. One long-standing unsolved challenge is the enantioselective aldol reaction of fluoroacetate to enable access to fluorinated analogues of medicinally relevant acetate-derived compounds, such as polyketides and statins. Herein we present fluoromalonic acid halfthioesters as biomimetic surrogates of fluoroacetate and demonstrate their use in highly stereoselective aldol reactions that proceed under mild organocatalytic conditions. We also show that the methodology can be extended to formal aldol reactions with fluoroacetaldehyde and consecutive aldol reactions. The synthetic utility of the fluorinated aldol products is illustrated by the synthesis of a fluorinated derivative of the top-selling drug atorvastatin. The results show the prospects of the method for the enantioselective introduction of fluoroacetate to access a wide variety of highly functionalized fluorinated compounds.

Polyene cyclizations offer rapid entry into terpenoid ring systems. Although enantioselective cyclizations of (E)-polyenes to form trans-decalin ring systems are well precedented, highly enantioselective cyclizations of (Z)-polyenes to form the corresponding cis-decalins have not been reported. Here, we describe the first application of iminium catalysis to the initiation of polyene cyclizations. Ethyl 1,2-diazepane-1-carboxylate catalyzes the cyclization of polyenes bearing enal initiators. Moreover, chiral bicyclic hydrazides catalyze the cyclizations of (Z)-polyene substrates to form cis-decalins with enantioselectivities of up to 97:3 er. DFT calculations suggest the catalysts promote the reaction by stabilizing positive charge as it develops during the bicyclization.

We report an extensive ?heteroarene scan? of triazine nitrile ligands of the cysteine protease human cathepsin L (hCatL) to investigate pi-stacking on the peptide amide bond Gly67?Gly68 at the entrance of the S3 pocket. This heteroarene???peptide bond stacking was supported by a co-crystal structure of an imidazopyridine ligand with hCatL. Inhibitory constants (Ki) are strongly influenced by the diverse nature of the heterocycles and specific interactions with the local environment of the S3 pocket. Binding affinities vary by three orders of magnitude. All heteroaromatic ligands feature enhanced binding by comparison with hydrocarbon analogues. Predicted energetic contributions from the orientation of the local dipole moments of heteroarene and peptide bond could not be confirmed. Binding of benzothienyl (Ki=4 nm) and benzothiazolyl (Ki=17 nm) ligands was enhanced by intermolecular C?S???O=C interactions (chalcogen bonding) with the backbone C=O of Asn66 in the S3 pocket. The ligands were also tested for the related enzyme rhodesain.
