Studies on fluorine containing condensation polymers are limited compared to that of fluorine containing addition polymers. In this report, highly fluorinated aromatic polyesters were synthesized by a polycondensation reaction of tetrafluorophthalic anhydride with ethylene glycol. Viscosity, solubility, thermal properties and crystallization behaviors of fluorinated polyesters were investigated using IR, 19F NMR, DSC, GPC, polarized optical microscope and rheometer. The fluorinated polyester is insoluble in most organic solvents, such as acetone, ethyl acetate, chloroform, THF, and trichloromethane. However, it is soluble in highly polar solvents, such as dimethylsulfoxide, dimethylformamide and dimethylacetamide. The fluorinated polyester (o-PETF) is a crystalline polymer with a crystallization enthalpy of 35.2 J/g and a broad crystallization temperature range from 54 to 130 C. Average crystalline growth rate is 4.2 mum/min at 110 C in the preliminary 30 min. Spherulite growth was observed at the temperature when the dendrites begin to melt. Crystallization property of o-PETF may be ascribed to the higher mobility of fluorinated polyester chains and dipolar contribution of carbon-fluorine bonds.

Licochalcone A (I), isolated from the roots of Chinese licorice, is the most promising antimalarial compound reported so far. In continuation of our drug discovery program, we isolated two similar chalcones, medicagenin (II) and munchiwarin (III), from Crotalaria medicagenia, which exhibited antimalarial activity against Plasmodium falciparum. A library of 88 chalcones were synthesized and evaluated for their in vitro antimalarial activity. Among these, 67, 68, 74, 77, and 78 exhibited good in vitro antimalarial activity against P. falciparum strains 3D7 and K1 with low cytotoxicity. These chalcones also showed reduction in parasitemia and increased survival time of Swiss mice infected with Plasmodium yoelii (strain N-67). Pharmacokinetic studies indicated that low oral bioavailability due to poor ADME properties. Molecular docking studies revealed the binding orientation of these inhibitors in active sites of falcipain-2 (FP-2) enzyme. Compounds 67, 68, and 78 showed modest inhibitory activity against the major hemoglobin degrading cysteine protease FP-2.

Notably, compound 5m (1 muM) could significantly sensitize four strains to immune clearance and could effectively attenuate the virulence of three strains in vivo. Moreover, 5m was determined to be a weak antifungal reagent (MIC > 16 mug/mL). Combined with good oral bioavailability (F = 42.2%) and excellent safety profiles, these data demonstrate that 5m may be a good candidate for the treatment of MRSA infections.
