Microglia can be activated to become the classic phenotype (M1) or alternative phenotype (M2), which play an important role in regulating neuroinflammatory response and tissue repair after ischemic stroke. CD21, a novel phthalide derivative, is a potential neuroprotectant against ischemic brain injury. The present study further investigated the effects of CD21 on post-ischemic microglial polarization and the underlying mechanisms. Transient middle cerebral artery occlusion (tMCAO) was used as a mouse model of ischemic stroke, while BV2 cells stimulated with conditioned medium collected from oxygen-glucose deprivation-treated HT22 cells were used in in vitro ischemic studies. The current results showed that CD21 dose-dependently and significantly improved neurological outcomes in tMCAO mice. Biochemical analyses revealed that CD21 decreased the expression of M1 phenotype markers (CD86, interleukin-1 beta and inducible nitric oxide synthase) and increased the expression of M2 phenotype markers (CD206, interleukin-10 and YM1/2) in both ischemic brain tissues and BV2 cells. Meanwhile, CD21 decreased the production of proinflammatory cytokines (interleukin-1 beta, interleukin-6 and tumor necrosis factor-alpha), promoted the release of the antiinflammatory cytokine (interleukin-10), and enhanced the phosphorylation of adenosine 5′-monophosphate-activated protein kinase (AMPK) in ischemic brain tissue and BV2 cells. Furthermore, the AMPK inhibitor (compound C) reversed these effects of CD21 in BV2 cells. These findings indicate that CD21 alleviates post-ischemic neuroinflammation through induction of microglial M2 polarization that is at least in part medicated by AMPK activation, suggesting that CD21 may be a promising candidate for protecting against ischemic brain injury.

Cnidium officinale Makino is an important medicinal plant of oriental clinics and is considered as the main source of phthalides, polyphenols, and flavonoids. However, there is no available report regarding the effect of different light colors on the secondary metabolites composition of C. officinale. In this study different light (dark, white, blue, red and red: blue) conditions were arranged to raise callus on MS medium containing 0.5 mg.L-1 of each 2,4-D and BAP. Callus grown in dark condition showed maximum (2.0 g) fresh weight with lower total phenolic and flavonoids contents. Also, in dark condition callus faced higher catalase (CAT) and guaiacol peroxidase (GPX) activities to avoid free radicals. Mix (red: blue) light condition favored the synthesis of phenolics and flavonoids in callus at the cost of higher ascorbate peroxidase (APX) and superoxide dismutase (SOD) enzymes expression. However, DPPH free radical scavenging activity was less variable among the samples from the different light conditions. Interestingly, the HPLC profile showed higher (28.3 mu g.g(-1) DW) phthalide accumulation in dark grown-cultures. Compared to other light conditions, 3-butyledinephthalide accumulation was higher (0.43 mu g-g(-1) DW) in white light-grown callus. These findings suggest that light conditions play an important role in the regulation of in vitro callus growth and synthesis of important medicinal compounds of C. officinale.

Plasma catalysis has been utilized in many environmental applications for removal of various hydrocarbons including tars. The aim of this work was to study the tars removal process by atmospheric pressure DBD non-thermal plasma generated in combination with packing materials of various composition and catalytic activity (TiO2, Pt/gamma Al2O3, BaTiO3, gamma Al2O3, ZrO2, glass beads), dielectric constant (5-4000), shape (spherical and cylindrical pellets and beads), size (3-5 mm in diameter, 3-8 mm in length), and specific surface area (37-150 m(2)/g). Naphthalene was chosen as a model tar compound. The experiments were performed at a temperature of 100 degrees C and a naphthalene initial concentration of approx. 3000 ppm, i.e., under conditions that are usually less favorable to achieve high removal efficiencies. For a given specific input energy of 320 J/L, naphthalene removal efficiency followed a sequence: TiO2 > Pt/gamma Al2O3 > ZrO2 > gamma Al2O3 > glass beads > BaTiO3 > plasma only. The efficiency increased with the increasing specific surface area of a given packing material, while its shape and size were also found to be important. By-products of naphthalene decomposition were analyzed by means of FTIR spectrometry and surface of packing materials by SEM analysis.

Fourteen new compounds including thirteen drimane – phthalide derivatives (fendlerals A – C, fendlerins A – D, fendlerols A – B, fendleric acids A – C, fendlerinine G) and one terphenyl derivative (fendleryl E) along with eight known compounds, fendlerinine A, rickenyls C – D, fendleryls C – D, atromentin, tetramethyl atromentin, and ( +/- )-microsphaerophthalide F, were isolated from the wood fungus Hypoxylon fendleri BCC32408. Compared with the prior work, the results indicated the agitation effect on the production of bioactive drimane – phthalides. The chemical structures were determined based upon spectroscopic analyses and the absolute configurations were verified by comparison of the ECD spectral data with the calculated ECD spectra of the related compounds. Compounds 1-3 exhibited antimicrobial activity against Plasmodium falciparum (IC50 4.15-4.39 mu M), Colletotrichum capsici (MIC 6.25-12.5 mu g/mL), and Bacillus cereus (MIC 1.56-3.13 mu g/mL). All tested compounds displayed broad cytotoxicity against cancerous (MCF-7, KB, and NCI-H187) and non-cancerous (Vero) cells.

A manganese-catalyzed oxidative kinetic resolution of cyclic benzylic ethers through asymmetric C(sp(3))-H oxidation is reported. The practical approach is applicable to a wide range of 1,3-dihydroisobenzofurans bearing diverse functional groups and substituent patterns at the alpha position with extremely efficient enantiodiscrimination. The generality of the strategy was further demonstrated by efficient oxidative kinetic resolution of another type of five-membered cyclic benzylic ether, 2,3-dihydrobenzofurans, and six-membered 6H-benzo[c]chromenes. Direct late-stage oxidative kinetic resolution of bioactive molecules that are otherwise difficult to access was further explored.

Five new phthalide dimers, chaxiongnolides C-G (1-5), the two enantiomers of riligustilide (6a and 6b) and tokinolide B (7) were isolated from the aerial parts of Ligusticum sinense Oliv cv. Chaxiong. The structures of 1-5 were elucidated through extensive spectroscopic analysis, and the relative configurations of compounds 2 and 3 were further confirmed by an X-ray diffraction experiment. The absolute configurations of 1-3 were assigned by comparing calculated and experimental ECD spectra. The ECD exciton chirality method was used to confirm the absolute configurations of compounds 4, 5, 6a and 6b. Compounds 1-3 are the first dimeric phthalides to be reported that are comprising a sedanolide unit. The compounds were evaluated in vitro for their cytotoxic activity against select human cell lines.

Aim:The development of effective PARP-1 inhibitors has received great enthusiasm in medicinal chemistry communities.Results:A new series of novel phthalazinone derivatives were designed and synthesized. Among these,B1andB16displayed more potent PARP-1 inhibitory activities than olaparib.B16gave an IC(50)value of 7.8 nM against PARP-1, and a PF(50)value of 3.4 in the sensitizing effect assay. Thein vivopharmacokinetic properties evaluation showedB16displayed insufficient oral exposure, and it was also not stable in rat blood.Conclusion:The results indicated that our design phthalazinone derivatives were potent PARP-1 inhibitors, and compoundB16was a valuable lead compound with siginificantin vitroefficacy, deseaving further optimization so as to develop anticancer drug candidate.

Flotation reagents, especially new chelating agents represented by alpha-nitroso-beta-naphthol, are the main components of cobalt mining drainage. This study reports the degradation of alpha-nitroso-beta-naphthol by simulated UVA-B (280-400 nm) activated systems using three common oxidants, hydrogen peroxide, sodium persulfate and potassium monopersulfate at a laboratory scale using a photoreactor. Parameters which can affect the degradation process were investigated and comparison of the degradation performance of the three systems were made. Based on the results, UVA-B/sodium persulfate system exhibited best performance towards the removal of alpha-nitroso-beta-naphthol with a lower cost of oxidant and energy consumption compared to the others. The removal efficiency was found to increase as the oxidant dosage and the UVA-B power increases. Only potassium monopersulfate could be activated by bicarbonate and chloride ions, and SO42- has insignificant effect on the removal efficiency of alpha-nitroso-beta-naphthol for all systems while NO3- inhibited the degradation of alpha-nitroso-beta-naphthol. In the UVA-B/hydrogen peroxide system, the hydroxyl radical had a leading role in the degradation of alpha-nitroso-beta-naphthol, while in the other two systems, the degradation of alpha-nitroso-beta-naphthol was mainly caused by the hydroxyl and sulphate radicals. Ten major intermediates from alpha-nitroso-beta-naphthol degradation in the three oxidation systems were identified by gas chromatography and mass spectrometry. In summary, this report could be a great input in developing UVA-B activated oxidants-based treatment technologies. The UVA-B/sodium persulfate system is strongly recommended for its consideration in the treatment of mine impacted wastewaters. (c) 2019 Elsevier Ltd. All rights reserved.

We report an efficient, catalyst-free, one-pot synthesis of (E)-N ‘-benzylidene-2-(hydroxymethyl)benzohydrazide derivatives by the one-pot reaction of phthalide, hydrazine hydrate and aldehyde. Advantages of this synthetic approach include mild reaction conditions and environmentally friendly nature, and the ability to provide a wide range of desired compounds in moderate to excellent yields.
