Aims: Partial PPAR gamma agonists attracted substantially heightened interest as safer thiazolidinediones alternatives. On the other hand, Wnt/beta-catenin antagonists have been highlighted as promising strategy for type 2 diabetes management via up-regulating PPAR gamma gene expression. We aimed at synthesizing novel partial PPAR gamma agonists with beta-catenin inhibitory activity which could enhance insulin sensitivity and avoid the side effects of full PPAR gamma agonists. Main methods: We synthesized novel series of alpha-phthlimido-o-toluoyl-2-aminothiazoles hybrids for evaluating their antidiabetic activity and discovering its mechanistic pathway. We assessed effect of the new hybrids on PPAR gamma activation using a luciferase reporter assay system. Moreover, intracellular triglyceride levels, gene levels of c/EBP alpha, PPAR gamma and PPAR gamma targets including GLUT4, adiponectin, aP2 were measured in 3T3-L1 cells. Uptake of 2-DOG together with PPAR gamma and beta-catenin protein levels were evaluated in 3T3-L1cells. In addition, molecular docking studies with PPAR gamma LBD, physicochemical properties and structure activity relationship of the novel hybrids were also studied. Key findings: Three of the synthesized hybrids showed partial PPAR gamma agonistic activity and distinct PPAR gamma binding pattern. These compounds modulated PPAR gamma gene expression and PPAR gamma target genes; and increased glucose uptake in 3T3-L1 and slightly induced adipogenesis compared to rosiglitazone. Moreover, these compounds reduced beta-catenin protein level which reflected in increased both PPAR gamma gene and protein levels that leads to improved insulin sensitivity and increased GLUT4 and adiponectin gene expression. Significance: Our synthesized compounds act as novel partial PPAR gamma agonists and beta-catenin inhibitors that have potent insulin sensitizing activity and mitigate the lipogenic side effects of TZDs.

Phthalides as the crucial core skeletons are found extensively in natural products and biological active molecules. Herein we disclose an asymmetric allylation of 3-hydroxyisoben-zofuran-1(3H)-ones with boron allylation reagents to construct chiral phthalide derivatives. The simple Bi(OAc)(3)/chiral phosphoric acid catalytic system proves to be efficient in this method, delivering the desired chiral 3-allylisobenzofuran-1(3H)-ones in good yields (up to 99%) and high enantioselectivities (up to 99.5:0.5 e.r.) under mild conditions. The large-scale reaction and diverse transformations of products to various scaffolds with potential biological activities render it more attractive. Moreover, the mechanism was preliminarily explored by control reactions, mass spectrometry, deuterium experiment, and DFT calculations.

A metal-free heterogeneous photocatalytic system has been developed for highly efficient benzylic C-H oxygenations using oxygen as an oxidant. This visible light mediated oxidation reaction utilizes graphitic carbon nitride (g-C3N4) as a recyclable, nontoxic and low cost photocatalyst. Mild reaction conditions allow for the generation of synthetically and biologically valued isochromannones, phthalides, isoquinolinones, isoindolinones and xanthones from readily accessible alkyl aromatic precursors in good yields. The heterogeneous nature of the g-C3N4 catalytic system enables easy recovery and recycling as well as the use in multiple runs without loss of activity. The synthetic utility of this green methodology was further demonstrated by applying in bioactive and drug valued target syntheses.

Air-stable iron carbonyl compounds bearing cyclo-pentadienone ligands with varying substitution were explored ,as catalysts in dehydrogenative diol lactonization reactions using acetone as both the solvent and hydrogen acceptor. Two catalysts with trimethylsilyl groups in the 2- and 5-positions, [2,S-(SiMe3)(2)-3,4-(CH2)(4)(eta(4)-C4C=O))Fe(CO)(3) (1) and [2,5-(SiMe3)(2)-3,4(CH2)(3)(eta(4)-C4C=O)]Fe(CO)(3) (2), were found to be the most active, with 2 being the most selective in the lactonization of diols containing both primary and secondary alcohols. Lactones containing five-, six-, and seven-membered rings .were successfully synthesized, and no over-oxidations to carboxylic acids were detected. The lactonization of unsymmetrical diols containing two primary alcohols occurred with catalyst 1, but selectivity was low based on alcohol electronics and modest based on alcohol sterics. Evidence for a transfer dehydrogenation mechanism was found, and insight into the origin of selectivity in the lactonization of 1 degrees/2 degrees diols was obtained. Additionally, spectroscopic evidence for a trimethylamine-ligated iron species formed in solution during the reaction was discovered.

Polyoxymethylene dimethyl ethers (PODE, with the formula of CH3O-(CH2O)(n)-CH3), as a promising environmentally benign diesel fuel or additive with an enormous potential in the reduction of soot and NIC, emissions, can be efficiently synthesized from methanol derivatives such as dimethoxymethane (DMM) and trioxymethylene (TOM), which requests a proper catalyst of high performance. In this work, the catalytic performance of graphene oxide (GO) in the synthesis of PODEn as well as its relation to the catalyst structure was thoroughly investigated. The results indicate that GO is an excellent catalyst in the synthesis of PODEn from DMM and TOM; a TOM conversion of 94.6% and selectivity of 86.0% to PODE2-8 are achieved under mild reaction conditions, which is superior to the majority of the state-of-the-art catalysts. Meanwhile, the active species and characteristic layer structure of GO sheets catalytically responsible for PODEn formation were discriminated through selectively removing certain surface functional groups, thermally annealing at different temperatures, and carefully comparing with a series of model compounds. The results illustrate that the superior catalytic performance of GO in the PODEn synthesis should be ascribed to a synergy between the surface sulfonyl, hydroxyl and carboxyl groups present on the GO surface and the unique layered structure of GO sheets, wherein the surface sulfonyl groups act as the main active sites. The insights shown in this work are beneficial to a deep understanding on the catalytic principle of GO and development of efficient catalyst for the synthesis of PODEn.

For the oxide-based photocatalysts, polyoxylic acid modification can affect their adsorption/desorption properties and further regulate photocatalytic reaction pathway, which is crucial for enhancing photocatalytic activity and selectivity. Herein, phosphoric acid modified TiO2 was synthesized by a facile impregnation-calcination method for photocatalytic degradation of gaseous o-xylene. According to FTIR, XPS, and EDS analysis, phosphoric acid was anchored on the surface of TiO2 successfully. The surface P atomic percentages for PT1 was 2.45%. The as-prepared phosphoric acid modified TiO2 (PTx) had a photocatalytic performance superior to that of commercial TiO2 and unmodified TiO2 (PT0), with increasing by 2.2 times at most. Interestingly, the phosphoric acid molecules strongly adsorbed on anatase TiO2 surface by forming H-O-2C and =O-Ti-5C chemical bonds according to the first principle calculations, which changed TiO2 surface properties (specific surface area and surface negative electrostatic field) and further improved adsorption and charge carrier separation and transfer, thus improving the photocatalytic activity. Additionally, according to the intermediates results, the relative abundance of intermediates shows obvious difference. Acetone was detected as the most abundant intermediates during o-xylene degradation for PT0, whereas that was o-methyl benzaldehyde for PT1, which could be ascribed to the difference of surface adsorption configuration of o-xylene. According to the temperature-programmed desorption (TPD) results, the surface modified by phosphoric acid could change the o-xylene adsorption configuration to favor its methyl oxidation (standing configuration), whereas the unmodified surface could be more favorable to the benzene ring-open reaction (lying configuration). This work will deepen the understanding of the relevance of surface modification and surface photocatalytic reaction, which also provides a feasible strategy to improve photocatalytic selectivity.

A great amount of insoluble phosphate in agricultural soils is not available for crops. Three strains of bacteria (Bacillus megaterium YLYP1 Pseudumonas prosekii YLYP6 and Pseudomonas sp. YLYP29) isolated from activated sludge and soil could efficiently solubilise tricalcium phosphate. In particular, the novel strain P. prosekii YLYP6 produced 716 mg L-1 of available phosphate within 6 days under the optimal culture conditions [20 degrees C, pH 7.9, inoculum size of 0.5% (v:v)] determined by response surface methodology. P. prosekii YLYP6 demonstrated efficient phosphate solubilisation in response to broad variations in pH (5-9) and temperature (15-35 degrees C). The phosphate solubilisation curves of the strains fit well with a first-order kinetic model (R-2 > 0.939), with a halt-life of 1.51-5.94 d for 5.0 g L-1 calcium phosphate. Continuous culture experiments combined with scanning electron microscopic observations and gas chromatography-mass spectrometry analysis revealed that 2,3-dimethylfumaric acid, gluconic and N-butyl-tert-butylamine that were produced by P. prosekii YLYP6 were responsible for phosphate solubilisation by supplying H+ ions and organic anions. Efficient phosphate solubilisation in actual soil by P. prosekii YLYP6 demonstrated the strong application potential to reduce the use of chemical P fertilisers and the resulting agricultural nonpoint pollution.
