The new compounds were screened as cyclooxygenase-1/ cyclooxygenase-2 (COX-1/COX-2) inhibitors and had analgesic and anti-inflammatory activities. The compounds 10a-d and 13a-f had the highest inhibitory activity on COX-2 selectivity, with indices of 99-90, analgesic activity of 51-42% protection, and anti-inflammatory activity of 68%-59%. The inhibition of edema for the same compounds, 10a-d and 13a-f, was compared with sodium diclofenac as a standard drug.

A comprehensive evaluation on pyrolysis behavior, kinetics, and primary volatile formation pathways of rice husk for application to catalytic valorization

Catalytic fast pyrolysis is a prospective and effective route to produce value-added products from rice husk (RH). Understanding the volatile compositions and its formation pathways as well as kinetics during RH pyrolysis is crucial and fundamental to regulate the quality of the target products since catalysts interact directly with primary volatiles. In this study, two pyrolysis coupled real-time volatile monitoring techniques (TG-FTIR and PyGC/TOF-MS) were employed to conduct comprehensive experiments for RH. The results showed that RH presented three mass loss and gaseous product evolution stages in the temperature range of 200 to 330 degrees C, 330 to 390 degrees C, and 390 to 600 degrees C, respectively. After speculating the formation pathways of the 24 main volatile species, it was indicated that 2,3-dihydro-benzofuran was the major product for hemicellulose, while 2-methoxy-4-(1-propenyl)-phenol was potentially a key active intermediate and very unstable during pyrolysis of lignin constituent in RH. In addition, pyrolysis of RH could be divided into two kinetic stages at cut-off point of 25% conversion. The first kinetic stage obeyed three-dimensional (D3) diffusion model and E-a was within 140-170 kJ/mol. In the secondary kinetic stage, Ea raised gradually to about 200 kJ/mol and pyrolysis reactions followed the second-order reaction model.

Novel potent (dihydro)benzofuranyl piperazines as human histamine receptor ligands – Functional characterization and modeling studies on H-3 and H(4 )receptors

Histamine acts through four different receptors (H1R-H4R), the H3R and H4R being the most explored in the last years as drug targets. The H3R is a potential target to treat narcolepsy, Parkinson’s disease, epilepsy, schizophrenia and several other CNS-related conditions, while H4R blockade leads to anti-inflammatory and immunomodulatory effects. Our group has been exploring the dihydrobenzofuranyl-piperazines (LINS01 series) as human H3R/H4R ligands as potential drug candidates. In the present study, a set of 12 compounds were synthesized from adequate (dihydro)benzofuran synthons through simple reactions with corresponding piperazines, giving moderate to high yields. Four compounds (1b, 1f, 1g and 1h) showed high hH(3)R affinity (pK(i) > 7), compound 1h being the most potent (pK(i) 8.4), and compound if showed the best efficiency (pKi 8.2, LE 0.53, LLE 5.85). BRET-based assays monitoring G alpha(i) activity indicated that the compounds are potent antagonists. Only one compound (2c, pK(i) 7.1) presented high affinity for hH(4)R. In contrast to what was observed for hH(3)R, it showed partial agonist activity. Docking experiments indicated that bulky substituents occupy a hydrophobic pocket in hH(3)R, while the N-allyl group forms favorable interactions with hydrophobic residues in the TM2, 3 and 7, increasing the selectivity towards hH(3)R. Additionally, the importance of the indole NH in the interaction with Glu5.46 from hH(4)R was confirmed by the modeling results, explaining the affinity and agonistic activity of compound 2c. The data reported in this work represent important findings for the rational design of future compounds for hH(3)R and hH(4)R.

A novel synthetic route toward vilazodone is described by using 4-cyanoaniline and 5-bromo-2-hydroxybenzaldehyde as starting materials, with an overall yield of 24% and 99% purity. Catalytic Carbonylative Double Cyclization of 2-(3-Hydroxy-1-yn-1-yl)phenols in Ionic Liquids Leading to Furobenzofuranone Derivatives

A catalytic carbonylative double cyclization method for the synthesis of furo[3,4-b]benzofuran-1(3H)-ones is reported. It is based on the reaction between readily available 2-(3-hydroxy-1-yn-1-yl)phenols, CO, and oxygen carried out in the presence of catalytic amounts of PdI2 (1 mol %) in conjunction with KI (20 mol %) and 2 equiv of diisopropylethylamine at 80 degrees C for 24 h under 30 atm of a 1:4 mixture of CO-air. Interestingly, the process was not selective when carried out in classical organic non-nucleophilic solvents (such as MeCN or DME), leading to a mixture of the benzofurofuranone derivative and the benzofuran ensuing from simple cycloisomerization, whereas it turned out chemoselective toward the formation of the double cyclization compound in BmimBF(4) as the reaction medium. Moreover, the ionic liquid solvent containing the catalyst could be easily recycled several times without appreciable loss of activity.
