We developed a method for the preparation of (diacetoxyiodo)-2-(N-alkylamido)benzene as an aniline-type hypervalent iodine(III). We also achieved direct cyclizations via C-H bond abstraction, such as the Hofmann-Loffler-Freytag reaction, a direct amination, and a direct lactonization, using the aniline-type hypervalent iodine(III) to obtain corresponding products in high yields.

Phthalides as the crucial core skeletons are found extensively in natural products and biological active molecules. Herein we disclose an asymmetric allylation of 3-hydroxyisoben-zofuran-1(3H)-ones with boron allylation reagents to construct chiral phthalide derivatives. The simple Bi(OAc)(3)/chiral phosphoric acid catalytic system proves to be efficient in this method, delivering the desired chiral 3-allylisobenzofuran-1(3H)-ones in good yields (up to 99%) and high enantioselectivities (up to 99.5:0.5 e.r.) under mild conditions. The large-scale reaction and diverse transformations of products to various scaffolds with potential biological activities render it more attractive. Moreover, the mechanism was preliminarily explored by control reactions, mass spectrometry, deuterium experiment, and DFT calculations.

Organocatalytic enantioselective intramolecular oxa-Michael reactions of benzyl alcohol bearing alpha,beta-unsaturated carbonyls as Michael acceptors are presented herein. Using cinchona squaramide-based organocatalyst, enones as well as alpha,beta-unsaturated esters containing benzyl alcohol provided their corresponding 1,3-dihydroisobenzofuranyl-1-methylene ketones and 1,3-dihydroisobenzofuranyl-1-methylene esters in excellent yields with high enantioselectivities. In addition, enantioenriched 1,3-dihydroisobenzofuranyl-1-methylene ketone could be obtained from the Wittig/oxa-Michael reaction cascade of 1,3-dihydro-2-benzofuran-1-ol.

Microglia can be activated to become the classic phenotype (M1) or alternative phenotype (M2), which play an important role in regulating neuroinflammatory response and tissue repair after ischemic stroke. CD21, a novel phthalide derivative, is a potential neuroprotectant against ischemic brain injury. The present study further investigated the effects of CD21 on post-ischemic microglial polarization and the underlying mechanisms. Transient middle cerebral artery occlusion (tMCAO) was used as a mouse model of ischemic stroke, while BV2 cells stimulated with conditioned medium collected from oxygen-glucose deprivation-treated HT22 cells were used in in vitro ischemic studies. The current results showed that CD21 dose-dependently and significantly improved neurological outcomes in tMCAO mice. Biochemical analyses revealed that CD21 decreased the expression of M1 phenotype markers (CD86, interleukin-1 beta and inducible nitric oxide synthase) and increased the expression of M2 phenotype markers (CD206, interleukin-10 and YM1/2) in both ischemic brain tissues and BV2 cells. Meanwhile, CD21 decreased the production of proinflammatory cytokines (interleukin-1 beta, interleukin-6 and tumor necrosis factor-alpha), promoted the release of the antiinflammatory cytokine (interleukin-10), and enhanced the phosphorylation of adenosine 5′-monophosphate-activated protein kinase (AMPK) in ischemic brain tissue and BV2 cells. Furthermore, the AMPK inhibitor (compound C) reversed these effects of CD21 in BV2 cells. These findings indicate that CD21 alleviates post-ischemic neuroinflammation through induction of microglial M2 polarization that is at least in part medicated by AMPK activation, suggesting that CD21 may be a promising candidate for protecting against ischemic brain injury.

We previously described a pH-sensitive phosphoramidate linker scaffold that can be tuned to release amine-containing drugs at various pH values. In these previous studies it was determined that the tunability of this linker was dependent upon the proximity of an acidic group (e.g., carboxylic acid or pyridinium). In this study, we confirmed that the tunability of pH-triggered amine-release was also dependent upon the pK(a) of the proximal acidic group. A series of 2-carboxybenzyl phosphoramidates was prepared in which the pK(a) of the proximal benzoic acid was predictably attenuated by substituents on the benzoate ring consistent with their sigma-values. The present study analyses the thermowell system for monitoring temperatures in a fixed bed catalytic reactor for the highly exothermic partial oxidation of o-xylene to phthalic anhydride. Particle-resolved computational fluid dynamics (CFD) simulations are presented for the cases of randomly-packed spheres inside cylindrical tubes with tube-to-particle diameter ratios (N) of 3.96, 5.96 and 7.99. Results are obtained for each N value for cases with inner thermowell radius to outer tube radius ratios of 0.0 (no thermowell), 0.05, 0.1 and 0.2. Comparisons of radial void fraction and axial velocity profiles, axial pressure drop and axial temperature profiles under reaction conditions are presented. The presence of the thermowell did change the mass and heat transfer and reaction inside and around the particles, especially at the bed center. The configurations of catalyst particles were rearranged, leading to changes in voidage, flow and temperature profile. Objective: The study aims at the derivatization of Phthalides and synthesizes 3-arylaminophthalides & 3-indolyl-phthalides compounds, and evaluates their anti-tubercular and antioxidant activities. The study has also intended to employ the in silica methods for the identification of possible drug targets in Mycobacterium and evaluate the binding affinities of synthesized compounds. Methods: This report briefly explains the synthesis of phthalide derivatives using ammonium chloride. The synthesized compounds were characterized using spectral analysis. Resazurin Microtiter Assay (REMA) plate method was used to demonstrate the anti-mycobacterial activity of the synthesized compounds. An in-silico pharmacophore probing approach was used for target identification in Mycobacterium. The structural level interaction between the identified putative drug target and synthesized phthalides was studied using Lamarckian genetic algorithm-based software. Results and Discussion: In the present study, we report an effective, environmentally benign scheme for the synthesis of phthalide derivatives. Compounds 5c and 5d from the current series appear to possess good anti-mycobacterial activity. dCTP: deaminasedUTPase was identified as a putative drug target in Mycobacterium. The docking results clearly showed the interactive involvement of conserved residues of dCTP with the synthesized phthalide compounds. Conclusion: On the eve of evolving anti-TB drug resistance, the data on anti-tubercular and allied activities of the compounds in the present study demonstrates the enormous significance of these newly synthesized derivatives as possible candidate leads in the development of novel anti-tubercular agents. The docking results from the current report provide a structural rationale for the promising anti-tubercular activity demonstrated by 3-arylaminophthalides and 3-indolyl-phthalides compounds.

Flotation reagents, especially new chelating agents represented by alpha-nitroso-beta-naphthol, are the main components of cobalt mining drainage. This study reports the degradation of alpha-nitroso-beta-naphthol by simulated UVA-B (280-400 nm) activated systems using three common oxidants, hydrogen peroxide, sodium persulfate and potassium monopersulfate at a laboratory scale using a photoreactor. Parameters which can affect the degradation process were investigated and comparison of the degradation performance of the three systems were made. Based on the results, UVA-B/sodium persulfate system exhibited best performance towards the removal of alpha-nitroso-beta-naphthol with a lower cost of oxidant and energy consumption compared to the others. The removal efficiency was found to increase as the oxidant dosage and the UVA-B power increases. Only potassium monopersulfate could be activated by bicarbonate and chloride ions, and SO42- has insignificant effect on the removal efficiency of alpha-nitroso-beta-naphthol for all systems while NO3- inhibited the degradation of alpha-nitroso-beta-naphthol. In the UVA-B/hydrogen peroxide system, the hydroxyl radical had a leading role in the degradation of alpha-nitroso-beta-naphthol, while in the other two systems, the degradation of alpha-nitroso-beta-naphthol was mainly caused by the hydroxyl and sulphate radicals. Ten major intermediates from alpha-nitroso-beta-naphthol degradation in the three oxidation systems were identified by gas chromatography and mass spectrometry. In summary, this report could be a great input in developing UVA-B activated oxidants-based treatment technologies. The UVA-B/sodium persulfate system is strongly recommended for its consideration in the treatment of mine impacted wastewaters. (c) 2019 Elsevier Ltd. All rights reserved.
