We report an efficient, catalyst-free, one-pot synthesis of (E)-N ‘-benzylidene-2-(hydroxymethyl)benzohydrazide derivatives by the one-pot reaction of phthalide, hydrazine hydrate and aldehyde. Advantages of this synthetic approach include mild reaction conditions and environmentally friendly nature, and the ability to provide a wide range of desired compounds in moderate to excellent yields. Flotation reagents, especially new chelating agents represented by alpha-nitroso-beta-naphthol, are the main components of cobalt mining drainage. This study reports the degradation of alpha-nitroso-beta-naphthol by simulated UVA-B (280-400 nm) activated systems using three common oxidants, hydrogen peroxide, sodium persulfate and potassium monopersulfate at a laboratory scale using a photoreactor. Parameters which can affect the degradation process were investigated and comparison of the degradation performance of the three systems were made. Based on the results, UVA-B/sodium persulfate system exhibited best performance towards the removal of alpha-nitroso-beta-naphthol with a lower cost of oxidant and energy consumption compared to the others. The removal efficiency was found to increase as the oxidant dosage and the UVA-B power increases. Only potassium monopersulfate could be activated by bicarbonate and chloride ions, and SO42- has insignificant effect on the removal efficiency of alpha-nitroso-beta-naphthol for all systems while NO3- inhibited the degradation of alpha-nitroso-beta-naphthol. In the UVA-B/hydrogen peroxide system, the hydroxyl radical had a leading role in the degradation of alpha-nitroso-beta-naphthol, while in the other two systems, the degradation of alpha-nitroso-beta-naphthol was mainly caused by the hydroxyl and sulphate radicals. Ten major intermediates from alpha-nitroso-beta-naphthol degradation in the three oxidation systems were identified by gas chromatography and mass spectrometry. In summary, this report could be a great input in developing UVA-B activated oxidants-based treatment technologies. The UVA-B/sodium persulfate system is strongly recommended for its consideration in the treatment of mine impacted wastewaters. (c) 2019 Elsevier Ltd. All rights reserved.

Aim:The development of effective PARP-1 inhibitors has received great enthusiasm in medicinal chemistry communities.Results:A new series of novel phthalazinone derivatives were designed and synthesized. Among these,B1andB16displayed more potent PARP-1 inhibitory activities than olaparib.B16gave an IC(50)value of 7.8 nM against PARP-1, and a PF(50)value of 3.4 in the sensitizing effect assay. Thein vivopharmacokinetic properties evaluation showedB16displayed insufficient oral exposure, and it was also not stable in rat blood.Conclusion:The results indicated that our design phthalazinone derivatives were potent PARP-1 inhibitors, and compoundB16was a valuable lead compound with siginificantin vitroefficacy, deseaving further optimization so as to develop anticancer drug candidate.

Utilizing a chiral bicyclic imidazole organocatalyst and adopting a continuous injection process, an alternative route has been developed for the efficient synthesis of chiral phthalidyl ester prodrugs via dynamic kinetic resolution of 3-hydroxyphthalides through enantioselective acylation (up to 99 % ee). The computational studies suggest a general base catalytic mechanism differing from the widely accepted nucleophilic catalytic mechanism. The structure analysis of the key transition states shows that the CH-pi interactions and not the previously considered cation/pi-pi interactions between the catalyst and substrate is the dominant factor giving rise to the observed stereocontrol.

A series of N-alkyl pyridinium salts were synthesized and employed as metal-free catalyst for the selective oxidation of methyl aromatic hydrocarbon with molecular oxygen. The electronic effect of the substitutes was found to be an important factor for the catalytic performance. With the introduction of electron-donating substitute -N(CH3)(2), the conversion of p-xylene and selectivity of p-toluic acid could be simultaneously increased. 1-Benzyl-4-N,N-dimethylaminopyridinium salt showed the highest catalytic activity, and 95% conversion with 84% of selectivity to p-toluic acid could be obtained for the selective oxidation of p-xylene. Several methyl aromatic hydrocarbons could all be efficiently oxidized with the reported catalyst at the absence of any metal species.

For the oxide-based photocatalysts, polyoxylic acid modification can affect their adsorption/desorption properties and further regulate photocatalytic reaction pathway, which is crucial for enhancing photocatalytic activity and selectivity. Herein, phosphoric acid modified TiO2 was synthesized by a facile impregnation-calcination method for photocatalytic degradation of gaseous o-xylene. According to FTIR, XPS, and EDS analysis, phosphoric acid was anchored on the surface of TiO2 successfully. The surface P atomic percentages for PT1 was 2.45%. The as-prepared phosphoric acid modified TiO2 (PTx) had a photocatalytic performance superior to that of commercial TiO2 and unmodified TiO2 (PT0), with increasing by 2.2 times at most. Interestingly, the phosphoric acid molecules strongly adsorbed on anatase TiO2 surface by forming H-O-2C and =O-Ti-5C chemical bonds according to the first principle calculations, which changed TiO2 surface properties (specific surface area and surface negative electrostatic field) and further improved adsorption and charge carrier separation and transfer, thus improving the photocatalytic activity. Additionally, according to the intermediates results, the relative abundance of intermediates shows obvious difference. Acetone was detected as the most abundant intermediates during o-xylene degradation for PT0, whereas that was o-methyl benzaldehyde for PT1, which could be ascribed to the difference of surface adsorption configuration of o-xylene. According to the temperature-programmed desorption (TPD) results, the surface modified by phosphoric acid could change the o-xylene adsorption configuration to favor its methyl oxidation (standing configuration), whereas the unmodified surface could be more favorable to the benzene ring-open reaction (lying configuration). This work will deepen the understanding of the relevance of surface modification and surface photocatalytic reaction, which also provides a feasible strategy to improve photocatalytic selectivity.

A practical, efficient, and mild hydrogenation of levulinic acid (LA) to gamma-valerolactone (GVL) under 1 atm H-2 was realized by single-sited 3D porous self-supported N-heterocyclic carbene iridium catalysts. Quantitative yields and selectivities were achieved at 0.02 mol% catalyst loading, and the catalyst could be reused for 9 runs without obvious loss of selectivity or activity.

Chemoselective hydrogenation of phthalic anhydride is regarded as the most promising route for producing downstream high-performance phthalide. A series of intermetallic cobalt-nickel silicide catalysts embedded in a carbon matrix (CoxNi2-xSi@C) with acid-tolerance prepared by microwave-assisted chemical vapor deposition have been investigated in this reaction system. Activity measurements show a remarkable positive synergistic effect, forming a volcano-shaped plot over the nominal Co metal fraction for CoxNi2-xSi@C catalysts with a peak at x = 1.5, which corresponds to a 4.5-fold enhancement of the yield of phthalide over Co2Si@C and a 2.0-fold enhancement over the Ni2Si@C monometallic silicide catalysts, which can be correlated with the interaction of cobalt-nickel and metal-silicon bonds. In addition, the cobalt-rich bimetallic silicide exhibits an equal activity to the reference noble metal catalysts (Au, Pd, and Pt) and a significantly higher activity than transition metal catalysts. Moreover, the bimetallic silicide as an intermetallic compound catalyst has demonstrated robust acid corrosion resistance compared to the corresponding metal catalysts.

Antagonists for the ATP-gated ion channel receptor P2X1 have potential as antithrombotics and for treating hyperactive bladder and inflammation. In this study, salicylanilide derivatives were synthesized based on a screening hit. P2X1 antagonistic potency was assessed in 1321N1 astrocytoma cells stably transfected with the human P2X1 receptor by measuring inhibition of the ATP-induced calcium influx. Structure-activity relationships were analyzed, and selectivity versus other P2X receptor subtypes was assessed. The most potent compounds, N-[3,5-bis(trifluoromethyl)phenyl]-5-chloro-2-hydroxybenzamide (1, IC50 0.0192 mu M) and N-[3,5-bis(trifluoromethyl)phenyl]-4-chloro-2-hydroxybenzamide (14, IC50 0.0231 mu M), displayed >500-fold selectivity versus P2X2 and P2X3, and 10-fold selectivity versus P2X4 and P2X7 receptors, and inhibited collagen-induced platelet aggregation. They behaved as negative allosteric modulators, and molecular modeling studies suggested an extracellular binding site. Besides selective P2X1 antagonists, compounds with ancillary P2X4 and/or P2X7 receptor inhibition were discovered. These compounds represent the first potent, non-acidic, allosteric P2X1 receptor antagonists reported to date.
