Utilizing a chiral bicyclic imidazole organocatalyst and adopting a continuous injection process, an alternative route has been developed for the efficient synthesis of chiral phthalidyl ester prodrugs via dynamic kinetic resolution of 3-hydroxyphthalides through enantioselective acylation (up to 99 % ee). The computational studies suggest a general base catalytic mechanism differing from the widely accepted nucleophilic catalytic mechanism. The structure analysis of the key transition states shows that the CH-pi interactions and not the previously considered cation/pi-pi interactions between the catalyst and substrate is the dominant factor giving rise to the observed stereocontrol.

The present study analyses the thermowell system for monitoring temperatures in a fixed bed catalytic reactor for the highly exothermic partial oxidation of o-xylene to phthalic anhydride. Particle-resolved computational fluid dynamics (CFD) simulations are presented for the cases of randomly-packed spheres inside cylindrical tubes with tube-to-particle diameter ratios (N) of 3.96, 5.96 and 7.99. Results are obtained for each N value for cases with inner thermowell radius to outer tube radius ratios of 0.0 (no thermowell), 0.05, 0.1 and 0.2. Comparisons of radial void fraction and axial velocity profiles, axial pressure drop and axial temperature profiles under reaction conditions are presented. The presence of the thermowell did change the mass and heat transfer and reaction inside and around the particles, especially at the bed center. The configurations of catalyst particles were rearranged, leading to changes in voidage, flow and temperature profile. A ligand-free iron-catalyzed method for the oxygenation of benzylic sp(3) C-H bonds by molecular oxygen (1 atm) using a thiyl radical as a cocatalyst has been developed. This transformation provides a facile access to amides, esters and ketones from readily accessible corresponding amines, ethers and alkanes. It features high regioselectivity, mild oxidative conditions and excellent functional group compatibility, providing good opportunities to the site-selective functionalization of complex molecules. Preliminary mechanistic studies suggest that this reaction may not undergo a benzylic cation intermediate pathway and the carbonyl oxygen atom in the products may be derived from molecular oxygen.

Microglia can be activated to become the classic phenotype (M1) or alternative phenotype (M2), which play an important role in regulating neuroinflammatory response and tissue repair after ischemic stroke. CD21, a novel phthalide derivative, is a potential neuroprotectant against ischemic brain injury. The present study further investigated the effects of CD21 on post-ischemic microglial polarization and the underlying mechanisms. Transient middle cerebral artery occlusion (tMCAO) was used as a mouse model of ischemic stroke, while BV2 cells stimulated with conditioned medium collected from oxygen-glucose deprivation-treated HT22 cells were used in in vitro ischemic studies. The current results showed that CD21 dose-dependently and significantly improved neurological outcomes in tMCAO mice. Biochemical analyses revealed that CD21 decreased the expression of M1 phenotype markers (CD86, interleukin-1 beta and inducible nitric oxide synthase) and increased the expression of M2 phenotype markers (CD206, interleukin-10 and YM1/2) in both ischemic brain tissues and BV2 cells. Meanwhile, CD21 decreased the production of proinflammatory cytokines (interleukin-1 beta, interleukin-6 and tumor necrosis factor-alpha), promoted the release of the antiinflammatory cytokine (interleukin-10), and enhanced the phosphorylation of adenosine 5′-monophosphate-activated protein kinase (AMPK) in ischemic brain tissue and BV2 cells. Furthermore, the AMPK inhibitor (compound C) reversed these effects of CD21 in BV2 cells. These findings indicate that CD21 alleviates post-ischemic neuroinflammation through induction of microglial M2 polarization that is at least in part medicated by AMPK activation, suggesting that CD21 may be a promising candidate for protecting against ischemic brain injury.

Methylation of sulfones, which have an alpha-CH bond, can be easily achieved via a one-step, Ru(II) catalyzed redox neutral reaction using methanol as a sustainable C1 building block. The reaction requires a stoichiometric amount of base and generates only water as a byproduct. A series of value-added methylated sulfones with various functional groups are produced under the reaction conditions from readily available substrates. Mechanism studies shows that a sulfone carbanion addition to an in situ generated aldehyde formed via catalytic dehydrogenation and subsequent catalyst mediated reduction of the alkene by hydrogen may be involved.

Organocatalytic enantioselective intramolecular oxa-Michael reactions of benzyl alcohol bearing alpha,beta-unsaturated carbonyls as Michael acceptors are presented herein. In order to develop novel chemotherapeutic agents with potent anticancer activities, a series of new 2,5-diaryl/heteroaryl-1,3,4-oxadiazoles were designed and synthesized. The structures of the new compounds were established using elemental analyses, IR and NMR spectral data. The compounds were evaluated for their anticancer potential on two standardized human cell lines, HT-29 (colon adenocarcinoma) and MDA-MB-231 (breast adenocarcinoma). Cytotoxicity was measured by MTS assay, while cell cycle arrest and apoptosis assays were conducted using a flow cytometer, the results showing that the cell line MDA-MB-231 is more sensitive to the compounds’ action. The results of the predictive studies using the PASS application and the structural similarity analysis indicated STAT3 and miR-21 as the most probable pharmacological targets for the new compounds. The promising effect of compound 3e, 2-[2-(phenylsulfanylmethyl)phenyl]-5-(4-pyridyl)-1,3,4-oxadiazole, especially on the MDA-MB-231 cell line motivates future studies to improve the anticancer profile and to reduce the toxicological risks. It is worth noting that 3e produced a low toxic effect in the D. magna 24 h assay and the predictive studies on rat acute toxicity suggest a low degree of toxic risks.

Flotation reagents, especially new chelating agents represented by alpha-nitroso-beta-naphthol, are the main components of cobalt mining drainage. This study reports the degradation of alpha-nitroso-beta-naphthol by simulated UVA-B (280-400 nm) activated systems using three common oxidants, hydrogen peroxide, sodium persulfate and potassium monopersulfate at a laboratory scale using a photoreactor. Parameters which can affect the degradation process were investigated and comparison of the degradation performance of the three systems were made. Based on the results, UVA-B/sodium persulfate system exhibited best performance towards the removal of alpha-nitroso-beta-naphthol with a lower cost of oxidant and energy consumption compared to the others. The removal efficiency was found to increase as the oxidant dosage and the UVA-B power increases. Only potassium monopersulfate could be activated by bicarbonate and chloride ions, and SO42- has insignificant effect on the removal efficiency of alpha-nitroso-beta-naphthol for all systems while NO3- inhibited the degradation of alpha-nitroso-beta-naphthol. In the UVA-B/hydrogen peroxide system, the hydroxyl radical had a leading role in the degradation of alpha-nitroso-beta-naphthol, while in the other two systems, the degradation of alpha-nitroso-beta-naphthol was mainly caused by the hydroxyl and sulphate radicals. Ten major intermediates from alpha-nitroso-beta-naphthol degradation in the three oxidation systems were identified by gas chromatography and mass spectrometry. In summary, this report could be a great input in developing UVA-B activated oxidants-based treatment technologies. The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and carbonucleophiles afforded poly(arylene alkenylene)s with moderate molecular weight in good yield. The reaction involves Mizoroki-Heck coupling, olefin migration via chain walking, and addition of the carbonucleophile to the resulting pi-allylpalladium species. The polymerization with a slight excess of nucleophile with respect to diiodoarene also proceeded to give the polymer without significant decrease in molecular weight in spite of the nonstoichiometric mixture of the monomers. The Pd-catalyzed three-component coupling polycondensation of diiodoarenes, nonconjugated dienes, and diimide also proceeded. The base used in the reaction is critical for yield and molecular weight of the product. The reaction using NaHCO3 afforded the product with low solubility, which can be explained by the high molecular weight of the polymer and/or the strong interaction of the electron donating dimethoxyphenylene groups and electron accepting diimide groups in the polymer.
